Genetic variability and potential effects on clinical trial outcomes: perspectives in Parkinson’s disease
Conclusions Our data support the hypothesis that within genetically unmatched clinical trials, genetic heterogeneity could confound true therapeutic effects as expected. Clinical trials should undergo pretrial genetic adjustment or, at the minimum, post-trial adjustment and analysis for failed trials

Five years with wrong diagnosis
I’m a 65 yo woman with medical background (pharmacist). First Parkinsons symptoms appeared more than five years ago: tremor both hands. I had some balance issues and abnormal gait. I always believed it was Parkinsons but the neurologist I first visited insisted it was essential tremor and treated with

High Dose B1 Thiamine HCL and Mannitol
My husband is currently taking Mannitol 20g a day, just trying since there have been reports saying it works to alleviate symptoms. We are keen to try HDT soon and wondering if we should continue Mannitol or just take B1 alone? Any comments is greatly welcomed! Sarah

PILLBOX - Identify or search for a pill (USA)
The FDA in the USA has a potentially useful tablet identifier tool. I have been aware of such tools for a long time but all too many appear to have some sort of restriction on access. (Which seems particularly unacceptable as identification can be an emergency requirement. And we don't always have someone

The efficacy of synthetic levodopa (without carbidopa) has been compared to that of natural levodopa (Mucuna) using rats with experimentally induced
parkinsonism
. The natural levodopa proved to be two times as effective at improving symptoms [32].

PD Paradigm shift: Brain-First versus Gut-First Parkinson’s Disease: A Hypothesis.
An important hypothesis about PD origins was recently published. The abstract conveys the rationale.(open access). IMO, researchers Per Borghammer and Nathalie Van Den Berge may have clarified and connected the diverse findings in PD and related alpha synuclein pathologies. I have long realized that

Change to Carbidopa Levodopa CR, dykinesia, neck dystonia
I recently saw my Neuro and had my prescription changed from C-L to an equivalent dosage combination of C-L and C-L CR (sustained release) to reduce off time. I went from three times 1-1/2 25/100 C-L pills per day to a regimen of one 25/100 C-L plus three 50/200 C-L CR pills per day. A presciption of

Important!! Canadian Dr. who prescribes Nilotinib Off Label
I have been following Georgetown University’s research with the use of Nilotinib, the 12 person trial, and the phase 2 75 double-blind recent trial. It appears to me that the drug is well-tolerated, safe if precautions are followed, and the results seem to indicate that it will arrest the progression

My Dad was finally diagnosed with PSP in December last year after being misdiagnosed with
Parkinsonism
. Since June 2018 he has lived at a Nursing Home but has steadily being going downhill. The hard part is that I live overseas so I don't get to spend nearly as much time as I would like with him.
