I have been invited to participate in a clinical trial (phase 2) at the Univ of WA, Fred Hutch cancer center: Vorinostat to Augment Response to 177 Lutetium-PSMA-617 in the treatment of patients with PSMA- low metastatic castration resistant prostate cancer: VALOR TRIAL. Have any forum people been in phase 1? Does anyone have first hand information on this beyond what is on the web? My PSA recently started climbing and is up to 30 now while on Jevtana and Carboplatin. What other treatments should I be asking about? Hormone therapy stopped working about a year ago after being on it for about 6 months. Taxotere worked well when I was on it a year ago. Six doses got my PSA down to .22. Is Pluvicto my last therapy option?

Here is link to the side effects of another trial with Vorinostat in CRPC patients. clinicaltrials.gov/study/NC...

Main side effects: Anemia (37.93%), Diarrhea (37.93%), Fatigue (86.21%), Platelet Count Decreased (51.72%), Anorexia (72.41%) and Hyperglycemia (37.93%).

If you combine Xtandi with Pluvicto it will have much fewer side effects and seems to work well. thelancet.com/journals/lano...

I didn't see Pluvicto alone vs Pluvicto with Enza in that study. Did I miss it?

Pluvicto is lutetium, which is the term used in the study

Yes. What I meant to say was that I didn't see a direct comparison of side effects where the study concluded there were less side effects of using Pluvicto with Enza vs. using Pluvicto alone. I did read a comparison of Pluvicto with Enza vs. Enza alone. But I'll read it through again.

I listed the side effects reported when using Vorinostat and consider these very severe. Therefore I recommend a different therapy, Pluvicto combined with Enzalutamide.

Enza will increase the PSMA expression and make Pluvicto more effective. It is not added to mitigate side effects. When I look at the side effects of Vorinostat I am convinced the Enza/Pluvicto combination will have much less side effects than Vorinostat.
