ASCO Genitourinary Cancers Symposium (Olaparib and Durvalumab in metastatic castrate-resistant prostate cancer (mCRPC))
Olaparib
is a PARP inhibitor [1]; Durvalumab is a checkpoint inhibitor [2].
Olaparib
[Lynparza] has had success in men with hereditary BRCA1 or BRCA2 mutations. As wonderful as newer drugs such as Zytiga are, resistance can lead to the emergence of cells that are very difficult to manage.

Adding PARP inhibitor (Olaparib) to the Standard Therapy (Zytiga/ADT/Prenosone)
I went to see him regarding adding Metformin to my list of medications (He's a well known expert - more on Metformin later), but the focus of the conversation turned into adding
Olaparib
to my existing treatment regime of Zytiga/ADT/Prednisone, because of my BRCA2 mutation.

Update - CT scan today, continuing Olaparib and 30 days in, of cannabis oil!
My current status (Jan 2017) is that the
Olaparib
/Lynparza is likely not working for me. My count & symptoms have continued to increase. Next step is likely Chemo in March I think... well that’s the oncologists plan anyway.

. Has anyone here tried this?

Good and bad news.
Bad news is that due to my anaphylactic shock with the carbo I can’t have
olaparib
for third line. He won’t give me cisplatin and he can’t or won’t desensitise me although I’ve asked him to look into that. He said they’d need a crash team and I’ve a 3% chance of dying.

Olaparib query
I'm worried that maybe cancer is there but is growing slowly because of
olaparib
. Now with the much smaller dose of
olaparib
cancer can start to grow faster. I am worried that
olaparib
maybe stops working for me.

Testing for BRCA Mutations.
He led on the first-in-man studies of
olaparib
that first demonstrated antitumour activity for PARP inhibitors in BRCA carrier patients. Professor de Bono has authored more than 200 peer-reviewed articles and several book chapters." Interesting discussion.

Disappointed and sad
He said if Rubraca is approved for maintenance by then, he will consider that one too but for now he thinks
Olaparib
. I knew it was a long shot but it has worked on others.
