Understanding mechanisms of resistance to Pirtobrutinib in Chronic Lymphocytic Leukaemia
Covalent Bruton Tyrosine Kinase inhibitors (BTKi) like ibrutinib,
acalabrutinib
and zanubrutinib have completely changed the treatment landscape for CLL.

Antibodies after third or fourth covid vaccine
Hi Everyone, Wondering if anyone developed initial antibodies, or increased previous antibodies after a third or fourth covid vaccine while on BTK inhibitors such as ibrutinib, or
acalabrutinib
? Also if you could provide the below information it could be helpful to our community. 1.

CoPay Assistance from LLS (Leukemia & Lymphoma Society) USA
I just started taking
Acalabrutinib
/ Calquence one week ago and the $2500 per month that my Medicare Part D does not pay is being covered by assistance from the LLS. And my 2020 tax return AGI Gross was well above what I thought any of the charitable agencies would approve.

Venetoclax: The Final Stretch……Do You Have A Uric Acid Problem
Later on, TLS became a more everyday issue especially during my time on Ibrutinib or
Acalabrutinib
when I think it became a much more acute problem.

BRUISING AFTER SUBCUTANEOUS INJECTION OF IMMUNOGLOBIN
Hello, friends, I have been on
Acalabrutinib
for 6 months now, and on Immunoglobin for 2 years. I wonder if any of you get bruising at the site of the injection. 2 injections per week. My belly is running out of space to do a new injection. Is there a way to minimize it? Thank you 🙏🏼

BTK inhibitor challenges by Dr. Nicole Lamanna
. - Nicole Lamanna, MD: With the advent of the BTK [Bruton tyrosine kinase] inhibitors and the fact that we’ve had many individuals on ibrutinib and now even
acalabrutinib
for many years, there’s no doubt that we’re starting to see some patients who develop resistant mutations to the covalent BTK inhibitors

There is also the risk I am using my back up drug too soon and there aren’t that many options after V and
acalabrutinib
. There are risks in not going after my leukemia while it’s down. Who knows what the long term side effects of
acalabrutinib
are?

Acalabrutinib and dry skin etc
I started treatment with
Acalabrutinib
in September and have been fine, no real side effects. My white blood count is now down to 24.1 and Heamaglobin in normal range. I just have a question for anyone else who is a
Acalabrutinib
user.

Alalabrutinib side effects after 18 months
I have been on
acalabrutinib
for 18 months. Initially I had the headaches, muscle and joint pain often associated with the drug. These symptoms dissipated after the first month. Now they seem to have returned, minus the headaches.

Changing from ibrutinib to acalabrutinib
From what I read from others these are all symptoms of ibrutinib side effects, and that switching to
acalabrutinib
may solve the problem with no loss in protection. Are there any ramp up, or periods in which all pills should be discontinued during the change over?

Fixed Duration A & O Phase 2 Study...anyone?
It's called: Phase 2 Time Limited Approach based on depth of Response to Front Line
Acalabrutinib
in combination with Obinutuzumab for CLL/SLL who achieve complete remission or partial remission with UMRD. Quite a mouthful!

Asking friends for pre-gathering precautions
My husband, currently on
Acalabrutinib
, would join us. We’ve asked that everyone be tested 2 days prior and not attend if they’ve traveled out of town (by air or otherwise) within the week before, nor if they’ve attended an indoor gathering of more than 25 people.

Advantage of Acalabrutinib over Ibrutinib?
What are the advantages of
acalabrutinib
? Disadvantages? Has anyone else made this transition? How did it go?

Still no antibodies because
Acalabrutinib
being a targeted drug affects the immune system. So no mixing still, all in good time. Stay safe and well.

CALQUENCE (Acalabrutinib) not working right after Covid vaccination?
I was on
Acalabrutinib
for over two years without any complications or side effects. I was vaccinated in January 2021. About 3 months after that I started to have issues with medication. I had to switch to Zanubritinb about month ago.
