Niclosamide phase 1 b trial showing more promise for CRPC than phase 1 trial
Eligible patients had progressing CRPC, adequate end-organ function, and no prior treatment with
abiraterone
or ketoconazole. Patients were treated with escalating doses of niclosamide/PDMX1001 and standard doses of
abiraterone
and prednisone.

Father recently diagnosed with metastatic PC
He will now start
Abiraterone
Acetate (and steroid). PSA has gone down from 500 to 42 after a month of the injection. The doctor has said the treatment will progress as follows - ADT along with Zytiga followed by possible chemotherapy/radiology if and after ADT fails.

Abiraterone Acetate
Dear all, Many thanks for your continuously support and sharing experience. Here I can share my concerns which previously I don’t know whom I could discuss with. As shared before, my dad is taking a half dose of Zytiga (500mg/day). We do not know if it is a new approach of not using full dose or because

Any input on Abraxane chemo?
My MO said on April 7 we’ll likely finish out
Abiraterone
ADT treatment next month in case it somehow works. If not, continue Lupron but take a “vacation” from
Abiraterone
and its side effects for about 3 months, and watch for a likely PSA rise.

PSMA scan
First Lupron injection Jan 11, and started
Abiraterone
Mar 9th. Plan is for Brachytherapy at UCSF then 25 External beam. Had a PSMA scan Monday with a PSA 0.8 Radiation Oncologist had this to say: I am out on vacation but I have looked at the report.

11-Ketotestosterone [11KT] (& 11-Ketodihydrotestosterone [11KDHT])

Abiraterone
targets adrenal-related androgens, so presumably 11KT isn't a problem for CRPC men on Abi?

Adaptive Theory for PCa - Part Two: Frameworks for treatment models, Eco-evolutionary Stability in mCRPCa, & AR is very adaptive
Gatenby, will be of more specific interest to PCa patients, as it used treatment outcomes of patients treated with
abiraterone
and focuses on the concepts that underpin AT when applied to mCRPCa in the clinic. It is also technical in nature and likely well above the pay-grade for most readers here.

Sartor on BAT and genomic profiling (excerpts)
But one of the things I did want to point out is if you want to come up with an enzalutamide response rate after
abiraterone
progression, what is it? About 30%? So, there is something, I think about 30% of these patients that may be manipulated.

The Magnitude Trial (and me, with scatological references)
This is a phase 3, large number international trial of the combination of
abiraterone
(abi + prednisone) with a newer PARP inhibitor (PARP-I), Niraparib (nira). It is run by Janssen/Johnson & Johnson who hold the patent for abi.

Dr Sartor raises questions about triplet therapy as a SOC for MHSPC. What was the take home message of PEACE I ??
While some have jumped on board and said the new regimen should be docetaxel plus
abiraterone
/prednisone plus ADT, Dr Sartor remains unconvinced...--SEE BELOW: https://www.practiceupdate.com/c/119674/67/11/?

Leuprolide and

+ prednisone for 2 years with most all the usual ADT SEs and PSA < 0.1 so far. A couple of months ago, I noticed significant but not severe swelling of my R calf and minor swelling of L calf.

Optimal Chemo-Hormonal Sequencing for MCRPC Maybe Taxotere - Zytiga - Jevtana - Xtandi
The study included two different groups: Group A (80 men) were given docetaxel chemotherapy (Taxotere) followed by one of the second-line hormonal therapies: either
abiraterone
(Zytiga) or enzalutamide (Xtandi) Group B (32 men) were given a second-line hormonal therapy then followed by Taxotere

Advice on dosage for Abiraterone
Oncologist is prescribing
Abiraterone
as 2nd line treatment now that I am castration resistant (mcrpc). Have read various info on taking 1000mg/day vs lower dosage of 250mg/day.
