Update on treatment with Olaparib (Lynparza)

I've been on Olaparib (Lynparza) for a few weeks. The original plan from Dr. Kwon at the Mayo was to combine Olaparib with Darolutamide (Nubeqa), but I've been unable to get the latter in Cyprus, where I live, and somewhat reluctantly I decided to go with Olaparib alone. PSA was still above 200 in recent blood tests. I'm concerned that Olaparib/Lynparza on its own may not produce much benefit. I've had pronounced swelling of legs and feet, lower-back and knee pain and difficulty walking. I recently started Lasix 20 mg daily, which has reduced swelling. I had to resume the antibiotic Augmentin for UT infection. I had stopped it and switched to Hiprex, but the infection returned. We then resumed Augmentin and it seems to have stopped the infection. I was able to discontinue it this past weekend (March 16). Bleeding from the UT appears to have stopped and I no longer have to use catheters.

I would be interested in others' experiences with Olaparib. My sense is that it may not work in my case, at least on its own) and I plan to return to the Mayo as soon as possible.

Many men have to discontinue PARP inhibitors due to toxicity.

I am BRCA2+. Your bio makes no mention of a gene mutation but generally this can make Lynparza more effective. I have been taking Lynparza for 10 months, along with Zoladex. I found 600mg a day overwhelming. Since October am on 400mg a day. Tolerable, blood counts ok and keeping the PCa at bay so far.

You may be able to fill the prescription at Mayo, when you are there, if it is not available in your country. It is very expensive without insurance.

I started Lymparza about 6 weeks ago. This is after about 5.5 years on Zytega. My BRCA2 status has prevented me from taking any successful therapy vacations. In year 5, my PSA was on a slow but steady increase. I am taking a "full" dose of Lymparza (600mg) and continuing my Zytega (Aberaterone Acetate).

After 6 weeks on Lymparza, I have had some very mild nausea and a marked increase in shortness of breath. I had my labs drawn today in preparation for a call with my Oncologist on Tues. My labs were predictably, a bit of a shit show. Red/white counts and HCT (Hematocrit).

I too am BRCA2 and took Lynparza by itself after Zytiga started to fail. Did not seem to help much. It made me feel “crappy” and had little appetite for weeks. PSA started to climb fast and switched to chemo.
