Hello everyone. I was diagnosed with Stage 3/4 ovarian cancer early in 2018. It was carboplatin sensitive, and I've now had two rounds of carboplatin (first with paclitaxel and then with doxorubicin), interspersed with bevacizumab, then olaparib. Unfortunately, it's back again.
I now have a choice of 'standard' carboplatin + gemcitabine, or a trial with a nanoparticle drug conjugate (which delivers chemo agent camptothecin) with the unalluring name of EP0057 (plus more olaparib). Besides the usual list of possible side effects, I haven't yet been able to find out what a fortnightly regime on the trial might be like e.g. does it allow you any 'good' days?! Has anyone else come across it? Thank you

EP0057 is manufactured by Ellipses Pharma, as you probably know.
Peer-reviewed publications stated: "Camptothecin-based drugs inhibit topoisomerase 1 (Topo 1), leading to destruction of DNA, and are currently being used as important chemotherapeutic agents in clinical anti-tumor treatment."
This is an international new trial that has very recently, last week, been launched in the UK (3 sites) so perhaps not many, if any, have been on it, as yet, to share their experiences.
Thank you. I do have the patient info sheet and have given it a good look over. Just have to decide now! One of the big questions for me arises from the fortnightly cycle - does this give you a chance to feel 'normal' for any time during the cycle, or is it all misery followed by recovery, followed by the next dose?
That's a pot of luck! No one knows what the precise answer is. You may very well tolerate it. It is an interesting trial. Personally, if I were to be offered the trial, I'd take it and if it didn't work, one can always revert back to chemo. I hope whatever decision you'll take it works best for you.
Thanks to everyone who replied to me about EP0057. Have started the trial and thought I'd give some very early feedback. Had my first infusion 4 days ago. On carboplatin-based regimes, I'd still be on all-day anti-nausea meds and probably lying on the sofa. I can't say I feel normal - am probably tired from dexamethasone wakefulness and I have the (non-Covid-19) cold that's going round and I've restarted the olaparib maintenance - so can't expect to feel normal. But I left out yesterday's midday anti-nausea meds and haven't taken this morning's either - and don't seem to be feeling sick yet. I would expect to get some olaparib nausea but I was managing that before without meds, so hope to do so again. Will report back on actual outcomes when I have some. Fingers firmly crossed.
