Hi well I’m back after a long break after previously having 2 BFN when we tried donor eggs with icsi. We’ve saved up again and decided to give it one last shot and this time the consultant recommended we try genetic screening before having any embryos implanted. Just wondered if there is anyone else about to go for this option or who has any recent success stories with this? Thanks and hoping it will be third time lucky 🤞🏻

Hi, I have 3 unsuccessful DE transfers. First two were from the same donor, third was a good grade from a new donor. We decided to PGTA test after the third negative transfer. We tested two of our remaining 4 embryos from the new donor and the results were 1 euploid and 1 mosaic. Transferred the mosaic which resulted in a BFP and baby boy arrived a couple of weeks ago. I'm glad we tested as I felt I needed to know if the issue was with the embryo or with me (unexplained infertility).
Hello and congrats! Out of interest can I ask why you decided the mosaic rather than the euploid? We are in the same situation and a bit lost!
Hi Miaow1975, we PGS tested our last 5 donor embryos and 4 were genetically normal. We transferred one of the normal ones and I am now 15 weeks pregnant. However, I also did an immune protocol for the first time for this cycle, as I have an overactive immune system. I am glad we did it as we had discussed it being our last cycle and it would have been a shame to inadvertently transfer the only abnormal embryo. Best of luck.

Hi Vassi, Thank you, we are taking one day at a time. I had immune testing with Professor Shehata as I kept on having early miscarriages and he put me on a personalised immune protocol, which was very strong as my results were so bad. It included Humira injections and hydroxychloroquine starting a few months before transfer, steroids, aspirin and intralipids 7 days before transfer (and then continued up to 12 weeks) and additional progesterone. A lot of clinics don’t buy into immune issues, but he gets very good results. Best of luck.
Hi Miaow1975 Sorry to hear you've not had success so far. It's so hard. We did pgta testing on our embryos from our second full ivf cycle. I'm glad we did as they led to us finding out that I have a chromosomal issue and we have been able to avoid using unbalanced embryos. I had 2 miscarriages during first round of ivf already and very likely abnormal embryos were the cause. Our first tested embryo that was normal didn't implant but that of course does happen. We did have genetic testing and a successful pregnancy but since then we have had a lot of abnormal embryos and I’ve been doing a lot of research over the weekend. There’s a lot of conflicting evidence - I.e. only a handful of cells are tested, embryos can self correct, sometimes the embryo sheds the bad cells and that’s what’s picked up by the PGTA test. It’s really made me reconsider.
In fact we had one embryo that was tested and came back as abnormal and retested under a different method and came back as normal.

Our clinic uses Igenomix and a few years ago they introduced Embrace testing which tests the fluid that the embryo secretes. They have to culture the embryos to day 6 to be able to do the embrace testing; it’s less invasive as you aren’t taking anything from the blastocyst. Anyway this showed several abnormalities and a year later we decided to do PGT and they came back normal.
