So my wife and I are in a disagreement about how many calories one of these half hour runs around week 6 burns. My Endomondo app I’m using says I’ve been burning 350 calories here or there abouts but she says it’s more like 100 cause that’s what the running machine in her works gym says. How can you tell what’s right?

I’d say week 6 is definitely more than 100.
As long as you've input your height and weight into your machine/app it gives you pretty accurate calorie burn
I use my fitness pal, Fitbit. Run keeper & strava they all give me around same answer going by my weight etc
I've had to put in a request to the developers of Runkeeper as the numbers don't go up high enough for my weight! 😃

My 28-30 minutes runs say between 400 and 500 calories in my runkeeper app
Well it will depends on how heavy you are - you burn 35-50 calories in half an hour just being alive without moving a muscle, so 100 is likely to be low as a total, but probably not far off as the additional calories burned from the exercise.
400-500 is more like the calories burned in an hour of intense HIIT
It depends on your weight, running efficiency, pace, gradient, temperature and a whole host of other variables.
Whichever is correct, neither is going to make you lose weight by itself.
I don't worry about calorie burn with running - weight training far better for that. Running is for your fitness and stamina, which in turn helps with your weight (HIIT) training, which helps with the calories burn and weight loss.
