I’m sorry I need a whinge
I know it’s not just me in this situation but I was told on Monday that, after a year,
Olaparib
is no longer working. I’m grateful I can have second line chemo rather than worse case, nothing to be done, which I do appreciate some of you do have to deal with. My sister wanted to talk me through it.

Should I reduce dose of Olaparib?
Because I am HRD I am being given Avastin 3 weekly and
Olaparib
300 mg twice daily. Unfortunately I am struggling with side effects. I get tired very easily. I have a very poor appetite and struggle to maintain my weight of 62kg.

Reversion mutation - re-challenging with a PARP no longer an option
After being on
Olaparib
PARP for 2 years, I recently had a Guardant liquid biopsy. It identified that my BRCA2 mutation has become a "[u]
reversion
[/u]" mutation, which means that PARPs will no longer be an effective treatment for me.

I recently had a Guardant360 CDx liquid biopsy that showed my BRCA2 inherited mutation, previously treated with
Olaparib
successfully for two years, is now classified as a reversion mutation. Prior to finding out this result, I had hoped that I could re challenge the use of
Olaparib
.

Tbh it has edged up every month on
Olaparib
(I have been on it since May 2021). i am really struggling with the idea that despite being BRCA 1 the maintenance treatment probably hasn’t really worked for me. Was it unrealistic to think I might get a longer remission?

So this buys time enough to really investigate the genomics and possible PARP inhibitor treatment (he mentioned
Olaparib
) And to hear someone of his reputation say 'You did right not to opt for the chemo - and you're a long way from needing Lutetium PSMA' was hugely reassuring.

Abiraterone/enzalutamide+PARP inhibitor better than abiraterone/enzalutamide alone for mCRPC, even without BRCA+

Reflection and thanks.
I had my last infusion of Avastin on Wednesday and am continuing with
Olaparib
tablets. I’m so thankful for the care I’ve received since diagnosis 19 months ago and we’ve successfully reduced the tumour, my CA 125 was 51, 4 weeks ago (it was 16985 in April 21!)

Good news update
. ❤️ She was diagnosed with OC Stage 4A in Dec, 2019 followed by surgery, pleural effusion draining, 6 rounds of chemo and on
Olaparib

when I first started
olaparib
4 months back, my Haemoglobin and platelets dropped significantly. I was asked to take 2 tabs (150mg each) twice a day. When haem dropped to 6.8, my doc reduced the dose to 1tab *2 and put me on iron supplement.

BAT treatment - when initiating?
Currently I have 0.1 and stable - using
Olaparib
. Should I start asap or wait until progression? Is std BAT to be preferred or utilize the new schemes current in trials. Any advice - I guess mant are in the same situation . (Chemonaive and prostate intact.)

Update on Mum- recurrence is appendix cancer not OC, olaparib and avastin stopped.
My worry is now how they will treat this recurrence and also they have stopped her targeted therapy- avastin and
olaparib
. She was 3 sessions away from completing the treatment… what will this mean for a recurrence of OC now, I don’t know.

I am having awful stomach pains and cramps since I started taking

a few weeks ago and would like to know about other people's experience of this . It is really affecting my quality of life and i don't know if I can manage to keep taking it.
