Hello, I am 36 yo with 50/50 condition. We are screening embryos pgd/pgs. I don’t have any known fertility issues as I have never tried for a baby and knew that I have a condition to screen for. I was wondering if many had success transferring first time euploid embryo of similar age? I am reading statistics and see really low percentages and it makes me really sad…

I don't have an experience with PGD test but I have only with PGS. I was 30 years old when I transferred first pgs normal embryo and it failed, it was a biochemical pregnancy. On second round we got pregnant with twins. At my clinic they are saying 65% of embryos are abnormal after the age of 35. My clinic's success rate per embryo transfer PGS tested is 91%, so a good chance to be successful. Good luck 🤞

Yes, I tend to believe about eggs for over 35yo, a lot of them don’t fertilise. May I ask if you don’t mind, why the first one failed transfer failed? Thank you and congratulations with twins ❤️❤️❤️

No, the eggs can be fertilised even 100% at any age but they are fertilised in an abnormal way, hence the results of 65% abnormal after age of 35. Hope this make sense.My transfer failed probably because my body didn't like "foreign bodies" and more likely was attacking the embryo. After taking steroids and immunosuppressants we been successful. It wasn't the embryo's fault, it was actually my body.

Hi, I'm also 36 and doing PGD and PGS (aka PGT-M and PGT-A) and very happy to tell you what I know.

Between the ages of 35-37 an average of 50% of the blastocysts you create are expected to be euploid. But, when you're dealing with small numbers (say 3-4 embryos from a single IVF cycle) the average number of euploids you get may be scewed, either below or above the average. To give you some context to this, my first IVF cycle we got just 1 euploid out of 5 embryos. However my experience is that over more than one IVF cycle we did get an average of 50% euploid embryos across all IVF cycles.

When you're also doing PGT-M that of course adds another layer of embryo attrition because you also need the embryo to be unaffected with the gene you have. However, provided you produce a good number of blastocysts and at least 1 or 2 of them are euploid it's a good sign that you'll be able to find one that doesn't have your gene. There is obviously an element of luck of the draw here, which is unfortunately inescapable.

In terms of success rates of transferring a euploid embryo, this will be very clinic dependent. From everything I've read I'd say your chances of getting a positive pregnancy test are high (roughly 70%), but your chance of a live birth will be more like 50% per euploid embryo transfer.

I have had one transfer of a good quality euplpid embryo which ended in a chemical pregnancy, so was on the wrong side of the 50% transfer odds at my clinic. Euploid embryos are very likely to get you to a live birth if you keep on transferring them, but it's good to be aware that they don't always work first time everytime. I say this because I know my expectations were set way too high for euploid embryos transfer and then when it didn't work first time it was a lot harder to deal with.

Thank you so much for your detailed response and amazing news that the second time it worked❤️ We had our first cycle unsuccessful of 5 embryos all had my gene but one and it was aneuploid (pgs tested only 2), now collected 10 from 2 cycles hoping to get better results ... If I may, why your first cycle was unsuccessful? Is it something before transfer I can do to make it work the first time? 😩

We've not had success yet, but we do have some euploid embryos now, so hoping we will be successful with a second or third transfer. That was really unlucky with your first cycle, I'm so sorry, that is so unfair! 10 embryos from 2 cycles will hopefully get you some good embryos after testing. Are you currently waiting for the test results on those?

We don't know why my first transfer didn't work unfortunately. Often it's impossible to know the exact reason. The embryo was good in that it was euploid and lining was also good. The only thing was that my progesterone was a little low on the day of transfer, but I'm not sure it was low enough for it to have made a huge impact. I think the most likely reason is that the embryo just didn't have what it takes to continue developing. I also tested before my official test date and that caused me a lot of distress because the lines weren't strong and part of me wonders whether the stress of that had an impact, but it's impossible to say for sure.

I've had two saline sonograms, microbiome and endometritis testing and everything has come back normal. I've also had a non-IVF pregnancy before (no live birth) so I know I've had full implantation before. I think a lot of the time, there's not much that can be done other than testing to rule out any potential issues and living healthily.

I see… the uncertainty is so difficult in the entire process, every step of the way (how many embryos… how many healthy. etc)… it adds when also before the transfer there is nothing certain and you basically don’t have power to improve or change anything..

We are in the situation when from second cycle we had 7 embryos good quality, from the third cycle we had 6, but 3 bad quality , therefore I said we only have 10 in total from two cycles as those 10 are good quality, but 3 are uncertain. So waiting to learn how many of those 3 are testable on day 5/6. Trying to stay realistic that 13 in total probably is unlikely… really difficult. Do you have a good euploid embryo number? What is your ivf reason?

The uncertainty is really difficult to deal with. The only thing that's helped me feel a little bit more in control is trying to understand enough about our situation and our treatment so that I can ask the doctors and nurses at my clinic the pertinent questions to try and ensure we're doing the tests and procedures that may help us the most.

10 embryos (potentially 13) is a very good number to send to testing, but I completely understand that you'll have that anxiety until you have the testing results back on those and you hopefully get some good ones. We now have 4 good quality euploid embryos after both PGT-A and PGT-M testing. We're very grateful to have gotten to this stage, but I don't think I'll be able to relax a bit more until one of those (hopefully!) sticks. We're having IVF solely to avoid passing down a gene I'm a carrier for.

I totally understand and I think having 4 of them is a crazy good base size and I am sure soon you’ll be a mom.I had few nerves lost while I was waiting for my results and so today they told me we have 11 embryos in total for testing from two cycles. We will wait 14 days for results. I have every hope in me. How many embryos have you had sent for testing? Thank you , sending positivity ❤️

I'm very grateful we have our 4, but I'm still anxious about whether one of those will lead to a live birth. Statistically I know we have good chances, but previous experience has made me question everything. That's great that you were able to send 11 in for testing. I'm keeping everything crossed you get good genetic results and I hope the next 14 days go quickly for you! We have sent a total of 12 embryos off for testing across 4 cycles. We lost 4 due to aneuploidy and 1 due to having my bad gene, 2 are mosaics which we'd only transfer if we were left with nothing else, 1 we transferred but was a chemical pregnancy and the 4 we have left are euploid.

Your result is a dream… to have only 1 affected by your gene (with autosomal dominant) and 4 due to aneuploidy is a very good result. Even those mosaics , I am sure would work well. I am praying to get more than one totally healthy.. from what i see , in my age, i think more than 1 or 2 healthy are highly unlikely.. so i consider you really lucky 🍀 what vitamins you take? Or anything else you do to improve the quality?

Very much hoping you get some healthy embryos from testing. We have averaged roughly 1.5 healthy embryos per cycle and I hope it's the same for you. Overall there's an average 50% chance of getting euploids for those in the 35-37 age range, but it can vary a lot between cycles. I've found it to be very 'win some, lose some', but you have a lot that are going to testing from two different egg retrievals, so I'd hope you'd definitely get something out of that.

We're keeping females who carry my gene but would be unaffected with the condition, which means we have a 3 in 4 chance of getting something to transfer after they've also tested euploid. But we've actually had 3 completely unaffected euploids overall.

I took a lot of vitamins for months before my cycles, although it's impossible to say if they helped or not. I took a prenatal, DHA omega 3, folate, vitamin C, D and E, ubiquinol (600mg), NAC, Alpha Lipoec Acid, NAD by Tru Niagen & pterostilbene. I also took ovasitol at the recommendation of my clinic, as they said it improved egg quality even for patients who don't have PCOS (I don't have PCOS). Husband also took a load of vitamins and we both cut out caffeine and alcohol for a few months.

There's not too many of us around doing PGT-M so it's always nice to speak to others going through a similar thing (even though it's really not nice having to go through this!). My next transfer is next month hopefully🤞

We did PGD for a 50/50 genetic condition. We achieved my daughter after three cycles. 🥰 No embryos in the freezer.

My sister in law also did PGD for the same condition. It worked first cycle for her (and she has an embryo in the freezer). 👍🌟🌈🤞

I had 5 from the first round, 2 from the second round and 1 from the 3rd.

We couldn’t use any from round 1, 1 from round 2, and 1 from round 3.

I thought we were out when we only got 1 day 5 embryo in our 3rd round, but it resulted in my daughter. 🥰
