My husband has only been on Lynparza for about five weeks. His las psa was 17.9 three weeks ago, today it’s 19.00. My heart feels so heavy. I feel so nervous. I feel as though we’re running out of options. He as I have mentioned has the CDK 12 mutation. Was on Zytiga that work for about four months. It worked in a sense that it kept his PSA stable they never really went down however it didn’t go up to much either. He has never done chemo. I’m thinking the doctors thinking immunotherapy next. If that doesn’t work I don’t know where to turn to next I am so devastated. Help guys please!!! 💔💔💔😭😭😭. Could really use some advice.

I have been on Lynparza for about 5 months. My PSA during that time has gone down from 2.5 to 2.0. My oncologist says it is normal to see such slow progress.

Interestingly, all but two of my 10 bone mets have shrunk by 30%. The other two have INCREASED by 30% or thereabouts - as per PET scans.

I have BRAC-2 which is the most favorable mutation for Lynparza to work.

Even with that advantage, it is weird how it shrinks some tumors while others tumors are increasing - in the same body!

Incidentally, most of the time I have been on 75% dosage - full dosage is too toxic.

I would not be concerned about a 1.1 increase in PSA. You might be pleasantly surprised by the scan results.

Zytiga did not work at all for me.

I’m concerned to the fact that. He is having some discomfort in different areas. What could that be?

I would suggest discussing chemotherapy with his doctor. The chemotherapies for Prostate Cancer have tolerable side effects for most.

Many people are afraid of chemotherapy because they have heard of or seen some very difficult experiences patients have had. But there is a big difference in the difficulty of chemotherapies. Docetaxel for Prostate Cancer, for example is on the easier end of the chemo spectrum. I did 6 cycles and it wasn't that difficult.

So it's something to consider and he only has to commit to trying one cycle. It may work for him.

Could you say something about the toxic effects for you on 75% and 100 % dosage? How fast did you notice the toxic effects?

Toxicity kicks in within days. Our bodies are all different, and some people can tolerate the toxicity (or don't care). If it works at 75% dosage, why put yourself through the misery?

In fact, I have just dropped to 50% dosage and will have the results of PSA in a few weeks and scans end of June.

I will report back end of June.

To answer Manohar11, I would do Chemo. It was very tolerable for me and took out every tumor in my body (bone and tissue - except the tumor in the prostate which it could only shrink).

It did not take out the circulating micro-mets, which is why I have bone mets two years after chemo.

Thanks! Another query if I may.... do you know if, contrary to what was the case for you with chemo, there is reason to hope that Lynparza will take out the micro-mets in addition to detectable mets?

Lynparza will stop working after about 12 - 18 months. Then the cancer starts its inexorable spread. There is no cure for us as yet (apart from a miracle cure).

I guess that´s how it is. Going for therapy after therapy, extending the time left for us.Positive to see that chemo worked for you for two years, despite what I understand is its reputation of not working for those with the BRCA2 mutation.

Don't be devastated, Slow PSADT isn't all bad. Must be working to hold it back... Chem looks like a treatment for the not to distant future if PSA keeps going up.
