Loss of PTEN

I have mCRPC since March. I am presently receiving chemo with taxotere and carboplatin. I have failed xtandi and zytiga. Lymph node tumor biopsy revealed adenocarcinoma, no NE. Genetic profiling revealed loss of PTEN. No other mutations.

My question is, what else can I be doing in regards to the PTEN? I am already on metformin 1 gm bid, resveratrol, quercitin, lycopene, pomegranate, turmeric/curcumin, and vit D3. I’ve read some on IC3 and DIM, but not currently using. All advice appreciated.

Not certain if it will help, but I take 4 g of metformin per day.

Also you might investigate Bipolar testosterone therapy.

What about lu177 and actinium pmsa treatments?

Those are possibilities after I finish chemo. More interested at this time in supplements that might be beneficial for the PTEN loss.

There are clinical trials with new meds which could be beneficial in patients with PTEN loss:

Indole-3-carbinol (I3C) may have some role in reactivating PTEN:

Look for clinical trials of PI3K/AKT pathway inhibitors. Unfortunately, some of the best trials have failed due to slow recruitment.

So from what I’ve read, the supplements that I should consider are I3C, DIM, and soy isoflavins with genistein. Is that correct? Anything in addition? Are these Life Extension products that are purchased on amazon, or other manufacturers? Are the dosages those listed on the bottle or something different? Thanks so much.
