My husbands genetic tests are back from Foundation One and he has a concerning mutation PTEN. He tested negative for BRAC, KRAS. If anyone has the same gene with any insight, we’d greatly appreciate your help. Thanks!

PTEN Loss gene and Advanced Prostate Cancer
It's very common with APC. Unfortunately, there are no known therapies for it.

Clinical trials are investigating carboplatin, and AKT inhibitors (Ipatasertib, afuresertib) for it.
PTEN loss is very common and the problem is it activates the P13K/AKT resistance pathway.

ncbi.nlm.nih.gov/pubmed/275...
Apigenin (APG; 4',5,7,-trihydroxyflavone) is a naturally occurring plant flavone abundantly present in common fruits and vegetables such as grapefruits, plant-derived beverages, parsley, onions, chamomile, oranges, tea and wheat sprouts. The most common source of APG consumed as a single ingredient in herbal tea is chamomile, prepared from the dried flowers of Matricaria chamomilla. Recently, APG has been increasingly recognized as a cancer chemopreventive agent. Numerous studies have explored the possible cancer chemopreventive effect of APG based on its potent anti-oxidant, anti-mutagenic, anti-inflammatory, anti-viral and purgative effects. The promising role of APG was summarized in various cancers including PCa.
Hi- There is some preliminary evidence that sulforaphane from broccoli family plants may target PTEN (see references below). Many forum members take sulforaphane containing broccoli supplements based on this and other evidence. You might consider adding in a sulforaphane containing supplement at higher doses or adding broccoli sprouts to the diet as they contain a high amount. In the absence of other ways to target PTEN, this may be a consideration.

There are clinical trials with AZD8186 which could work in patients with PTEN mutations:

They are recruiting at the MSKCC in NY, at the MD Anderson in Houston and at JHopkins in Baltimore.

Best of luck.
Good find. I went through all the clinical trials looking for P13K inhibitors, AKT inhibitors and M-tor inhibitors. I couldn't find any that were active and recruiting.
Paige,

My husband, entire family and I are incredibly grateful to all of you!
May I ask another question.. does anyone know if having PTEN loss effects Lu177 treatment?
