Latest acalabrutinib results and from ASH 2018: Dr. Mato on a new Triple Combination of Novel Drugs for CLL and Richter’s Transformation
Hi, Things don’t slow down much in the CLL world. And that is a good thing. Just makes for a lot of writing for me to keep up. In the last week we posted about the encouraging results in an acalabrutib trial and my interview with Dr. Mato at ASH 2018 about a new triplet therapy (Ublituximab (TG-1101)

How can I get acalabrutinib?
Some of you have tales about getting
acalabrutinib
offline and getting insurance to approve it. Can someone please explain this process to me so that I have more information when I see my doctor on Monday? Thank you so much. I am new to this forum and have learned so much in the past few days.

New Years Good News
Now, wonderful success with
acalabrutinib
. No adverse side effects to speak of. All is good! When I was first diagnosed, everyone told me that blood cancer research was moving fast and that new and amazing treatments were on the way. I didn't believe them at first. Now I do!

My Dana-Farber Trial Update
Started
Acalabrutinib
in August and had a couple of headaches over the first week. Called my care team. Tylenol basically stopped the headaches. They never came back. Zero side effects from Obinutuzumab. Today, I completed my first dose of Venetoclax at 400 mg after titrating up for a month.

Dr Wierda recommended me the clinical trial on
Acalabrutinib

Anyone taking calquence (acalabrutinib)?
My doctor wants me to take calquence (
acalabrutinib
). However, I am even afraid of that after my experience with imbruvica. I am leaning towards immunotherapy. Is anyone on calquence? If, is all going okay? Thanks, Rich

Successful start of second arm of Acalabrutinib, Obinutuzumqb, Venetoclax Trial
One month on
Acalabrutinib
knocked my nodes down to less than half and my WBC from 82,000 to 62,000. Started the Obinutuzumqb arm yesterday with the 1/10th dose, with side effects only from the Benedryl.

MUST READ for those on Ibrutinib/Imbruvica - concerning advice in pilot study on lower dosing - incl. danger of starting with a lower dose
BCR antagonists, likely ibrutinib,
acalabrutinib
, idelalisib, all inhibit their target kinase at a level that is dependent upon their concentration in the serum. Currently, most agents are dosed well above the necessary level to achieve 100% inhibition.

Not even a week on Acalabrutinib and...WOW!
Well, I've been in the Dana-Farber clinical trial for almost a week, starting with
Acalabrutinib
. This weekend, after four full days on it, my right ear started popping, like when changing elevation on a flight or as an infected sinus clears. I was wondering if this was coincidence.
