First, a look at a new mitochondrial enzyme target for those failing treatment. The process of ferroptosis can lead to cancer cell death and inhibit tumor growth, but a mitochondrial enzyme (DHODH) can block that process. Researchers have found that using brequinar, a drug being studied for possible use against CoVid-19 and AML--see below:
The study for targeting DHODH showed even greater efficacy when used with sulfasalazine, a drug with some efficacy against PCa. Brequinar should complete Phase II trials by June, and is a drug to watch in treating different cancers. A possible game changer drug. The article below:
Next, a clinical trial worth looking at for PCa patients that are oligometastatic is the Metacure trial. Being done in several locations, the trial looks at using ADT plus apalutamide and possibly abiraterone prior to removing the prostate. They do have a post prostatectomy cohort as well. The trial includes lymph node removal for suspicious nodes and SBRT for osseous lesions in an attempt to shoot for a cure.
Ferroptosis is a process that I have been looking at for a while. Sulfasalazine, an anti-rheumatic drug which decreases inflammation has been shown to be synergistic with high dose Vitamin C in killing PCa cells by increasing ROS in cells.
As Sulfasalazine inhibits a cystine-glutamate transporter, it can promote ferroptosis. Sulfasalazine is a cheap old drug that I keep in the back of my mind for possible use...
A positive response is a good thing... I keep my eyes open, but right now, just enjoying a break... People that are having problems should look at alternatives to SOC.....things like heme iron, sulfasalazine and high dose IV Vitamin C, etc... if nothing else then for add-ons to SOC... all IMHO...Have a good weekend....
You guys seem to be way ahead of me, as I think both of you have posted before on ferroptosis. When I saw this post go up, i looked down in front of my keyboard and there was a sheet of paper with the words "ferroptosis" and "glutathione" written in the center. I couldn't remember what caused me to make the two-word note, but it might have come from this paper?

Based on your articles, the 3 drug combo should be quite effective... it provides everything needed and could even be used with SOC hypothetically... The idea of finding old drugs to use with SOC is something that has interest to me...

Nothing like a 3 drug cocktail....just look at HIV.... I do wonder if someone might not "give it a go" and do n=1 and report... for someone stable on SOC, a chance to reduce tumor burden and possibly get to "undetectable"...
