Multi-nuclei embryos

Hi had anyone else had any experience with their embryos being classed as multi nuclei?

We have had 8 failed embryos now (1 was miscarriage) they have looked back at all our embryos that made it to day 5 blastocyst and most have multi nuclei in the cells which they think is causing the implantation failure.

Really just wondering if anyone has had this before or can offer any advice?x

I don’t have experience of this personally but it’s my understanding that the pro nuclei bit is relevant that first day after fertilisation and that what they are looking for is a 2 pro nuclei (2PN) fertilisation of the egg - ie a copy of dna from the mum and one from the dad. I have heard of a lady on another forum having a successful pregnancy with an embryo which stared out as 1PN - I would perhaps ask if you are seeing a lot of 3PN embryos then could you be a carrier for a trisonomy yourself - had you or partner done any karyotyping?

Hi thank you for your reply, our embryo's are all making it to day 5 blastocyst but with multiple (at least 3) nuclei in each cell, we have just had genetic testing done (£800 for 2 blood tests!😣), but the consultant has said they really have no idea of the cause or how to treat, the tests take 2 months to come back so just trying to think about what our options are

Do you know what your E2 levels were on trigger day? I’ve heard that high incidences of 3PN could be related to high E2 levels on that day?

No I've never been tested for anything to be honest, and I've never heard of that so will definitely look into it thank you, I don't ever have HCG for my trigger shot due to high AMH and OHSS, do you know if that makes any difference?

I think it’s more related to your E2 estrogen level than hcg trigger - if you have OHSS and a high AMH that would make sense as high E2 is associated with lots of follicles - what level of stims were you on? I’d be inclined to explore a low dose protocol (I had twins via natural modified IVF after having poor fertilisation and rubbish egg quality)

that's brilliant advice, thank you so much! I normally have buseralin as my trigger, 150mcg/75mcg on alternate days, on my first egg collection I had 36 follicles so they reduced it down but they have said they will go further down again next time, but then will run the risk of not producing enough mature eggs! it's such a fine balance!
