Suggestions after second failed implantation

Hi everyone,

I’ve spoken with my clinic and doctor and as I suspected they said it is just bad luck the second transfer failed as well. I was wondering if you could please give me some suggestions on what I could ask the doctor? Or if anyone else has had failed implantation was there any other testing that they could do either on my uterus or the donor eggs we have remaining?

I have had a hysteroscopy which they did not find anything. I have also had implantation cuts. My husbands sperm had been tested and it came back with great results. The donor eggs we had all made were grade 5aa blastocyst. We have three remaining.

The doctor would like me to try again on the next cycle, to take advantage of the implantation cuts while they are still there.

Any insight or feedback would be great.

Hey hun, I have had two failed Implantation aswell. I went on to have my NK cells testsed at covernty and Warwick hospital and they came back slightly high. So this next cycle I am going to take steioids to see if this aids implantation. Fingers crossed it does help! Sorry for your cycles not working. Take time to heal and when you feel ready try again xx

I will ask about the NK cells but I know was given prednisone which I think is used for treating NK cells?

How do they test for NK cells? Is it a blood test?

They took a biopsy of my womb. Again yeah those are the tablets I am going to be put on! I also had a scratch done on day 21. Let's see if these changes work! X

We had some biopsies taken at same time as hysteroscopy, consultant cut away what he called thickened scar tissue. Biopsies came back clear but my bloods have come back with a few clotting issues so on clexane for next round and also having intralipids before transfer this time to help with slightly raised nk cells. I have had two failed transfers with my OE cycles and thought we had cracked it moving to DE. We transferred 2 hatched blastocysts and our consultant was convinced this would be our time but no. Hoping the changes help for next round. Perhaps it's just bad luck, nobody really knows!xx

Thanks Cinderella, I know I had some tissue cut away as well during the hysteroscopy. And both of these failed rounds I have been taking the clexane injections for clotting. I will ask about the NK cells but I know was given prednisone which I think is used for treating NK cells? Maybe I need a higher dosage?

I hope both our rounds are ours next time. I will have my fingers crossed for us both xx

My consultant wasn't keen for me to do the prednisolone. I'm doing intralipids only....bit unsure of the reason. Hoping we've just had really bad luck!!xx

I’m not sure what intralipids are? I will have a look and see what the difference is and see what my doctor says. Thank you though for your support and the information xx

I posted about intralipids if you look back my posts. I've no idea of the science behind it as it's basivally an IV of soya oil & eggs but seems to work for some. lianm8 used this on both of her successful rounds! You're welcome, it's good to know we're all there for each other!!🤗xx

Hi Cinderella, hope you’re well? Did you get the level 2 bloods done? I recall you were thinking about exploring the nk cells testing. How did you get on? I think nk cell testing will be my next step following my 3rd miscarriage. X

Not sure what level 2 bloods are?! My clinic have done so many bloods and have been very thorough! Found clotting issues & no cell issues. So clexane and intralipids for next round. I really hope you you find answers for your miscarriages!xx

Chicago level 2, nk cells I believe. Level 1 tests cover the blood clotting issues x

I had mine done altogether with my hysteroscopy which includes uterine biopsies & level 1&2 bloods. It came to €2000 so not sure how much it would have been separately.xx

Double transfer, they don’t seem to want to stick :( We have three frosties left now. So will do two again and then will have one left

My clinic has suggested a huge list of bloods to check for possible causes. In uk normally only done after at least three failed or miscarriages xxx

Protein c and s lupus anticoagulant,

Anti thrombin 111 homocysteine, anti b2 glycoproteins IgG and igm

Anti cardiolpin IgG IgM iga

Cykoteines th1/th2

Lymphocytes sub population

Prothrombin gene factor 11

Detection of 2 mutations gene cftr and

Detection of polymorphism mthfr

Sorry to hear this. It is very hard. Have you thought about using the embryo glue? According to the HFEA it shows promising results. We always use it xx

Thank you! I’m sending the consultant an email with questions and will definitely see if we can use embryo glue.

Hi Smang, I’m not sure if it’s relevant or not but following my third miscarriage I am going to look into natural killer cell testing and I have read that it can also impact implantation. Quite a controversial subject but could be something to look at? Good luck! X

Hi. I have had 2 failed cycles as well. I timed both with the bcp pill and I don't next time I have a transfer. I feel it just doesn't suit my body. There is a bacterial infections test you can do at locus medicus in Greece. If u search it u can send your samples via post and then they provide the results via email and if diagnosed advise a way forward. I hear that bacterial infections can cause embryos not to stick or make if further. Yes nk cells are a must to test. I'm hoping to go to Coventry to check mine but only after I have had a hysto. Dr s in London usually puts people with elevated nk cells on 20mg steroid daily and other meds such as resveratrol high omega fish oil and vitamin d. Hope this helps.

Thanks for your message, I’ve had the bacterial infections test as my clinic is in Athens also, they took a sample when I did my hysto. Interesting though about the vitamin D I started taking last month and that was because my GP suggested it. So hopefully for next round it would have kicked in.

My clinic has put me on resveratrol, omega fish oil, vit D and but C too Smang .xx

I guess any thing is worth a shot isn't it hun. For my next round the only thing I won't be having is the LIT otherwise I'm trying it all.
