I continue to believe that research into treating Covid-19 may provide benefits to Parkinson's patients and vice versa.

Indeed could be some similar antagonists between the Covid and PD! Some time ago Roche purchased a biotech named Inflammazone as Roche must have seen something in their makeup that might help to bring a treatment for PD and other diseases. Wondering where that is all at and if Roche is involved in looking into ties between Covid and PD?

Research paper: SARS-CoV-2 drives NLRP3 inflammasome activation in human microglia through spike protein

nature.com/articles/s41380-...

Melatonin has shown benefit for both diseases and it inhibits NLRP3 imflammasome activation while very significantly reducing the elevated oxidative stress levels seen in both diseases.

dovepress.com/melatonin-att....

Here is a relevant quote :

' Once the NLRP3 inflammasome is activated, it cleaves pro-IL-1β into mature IL-1β and promotes the release of ASC specks. However, melatonin can negatively regulate NLRP3 inflammasome activation via the SIRT1-dependent pathway and protect dopaminergic neurons in Parkinson’s disease. '

In people with PD, melatonin returns oxidative stress levels to healthy control levels :

ncbi.nlm.nih.gov/pmc/articl...

Here is a relevant quote :

' In conclusion, melatonin administration was effective in reducing the levels of oxidative stress markers and restoring the rate of complex I activity and respiratory control ratio without modifying membrane fluidity. This suggests that melatonin could play a role in the treatment of PD. '

See graph at bottom of this post to see exactly how much melatonin reduced oxidative stress markers in PwP at 50 mg/day for three months. As you can see, it is essentially at healthy control levels.

Melatonin offers similar effects in SARS CoV-2 patients :

pubmed.ncbi.nlm.nih.gov/343....

Here is a relevant quote :

' Based on the findings of the current study, it is found that melatonin can be effective as a medicinal supplement in decreasing the inflammasome multiprotein complex and oxidative stress along with beneficial impacts on lung cytokine storm of COVID-19 patients. '

The body produces melatonin naturally, but that production decreases sharply with age and in people with PD, it declines even more than in healthy controls.

Art, are you still taking melatonin? How many mg per day. I just increased to 80.

I have now completed one year at 106 mg+/day and just completed my second year at 120 mg+/day. I am now debating on whether to go higher or stay at 120 mg+/day. So two consecutive years total at over 100 mg/day.

I apologize if you have already answered this, but where do you buy it? I buy 10 mg pills at Walmart, but I would like to take fewer pills with more mg.

If you are looking for the largest for oral use, these are a few of the ones I have seen in 60 mg capsules :

These are the highest I have seen as a suppository :

Hi Jim caster ~ this is the 60 mg melatonin that I give my HwP. It has a couple of other things in it. Also, 60 mg was too much for him so I alternate ~ 40mg one night ~ 60 mg the next night. I couldn’t find 50 mg’s at the time.

Hopefully the link works

amazon.com/Carefast-Melaton...

If the link doesn’t work the brand is Care Ease Melatonin Plus on Amazon.

Have a fabulous day!

I buy bulk melatonin powder. You can make your own suppositories :)

Last I knew, Art was taking 106 mg daily. This post has comments with lots of information regarding melatonin.

If you are asking about melatonin and PD, 10mg/ day and 50 mg/day have shown benefit in PwP.

The highest longer term dose I have seen in humans was 3000 mg/day for 6 months.

pubmed.ncbi.nlm.nih.gov/279...

A relevant quote from the study "

' One group took myo-inositol (myo-Ins) (2 g twice a day) and the other group took 2 g/d myo-Ins plus 3 g/d melatonin before sleeping. '

I checked with Professor D'Anna, the first author in that paper about the 3000mg dose and it's a typo, they left an "m" out: it should read 3mg.

That study is not new, it seems odd that they never corrected such a very significant error. From 3 grams to actually 3 mg, wow!

Im on 20mg. And sleep like a baby with lots of interesting dreams that i write and learn to decode as soon as i get up. If i could lower the amount of supplements ( trying with herbal tinctures) i probably would be able to afford a higher dose. I think it makes a difference on my functioning...
