Graph showing levodopa levels during the day: on/off and dyskinesia
Some time ago I wrote an app that models how
levodopa
levels vary as a result of the medications that we take. This is based on pharmacokinetic values such as the half-life of a drug.

DBS and Sinemet
I had the DBS surgery last October. After DBS, Sinemet made my PD symptoms worse...especially balance (It's like I am constantly falling forward and my legs are trying to keep up). I told my neurologist about it and she thought is odd that Sinemet would do that. I feel better without the Sinemet.

shuffling feet
I take 3 tablets of carbo /

, 25 - 100mg, 3 - 4 times a day, I never take C/L after 5pm and I take Amantadine., 3pm, 5pm &7pm which controls the dyskinesia. I get plenty of exercise, but the feet shuffling is something new, any suggestions?

Is high dose thiamine therapy effective for people not taking levodopa?
If I understand correctly this therapy "is a co-adjuvant therapeutic measure to use with
levodopa
therapy". Does it mean this therapy won't have any effect on my friend not taking
levodopa
? They are taking amantadine and pramipexole. Can B1 therapy help?

opicapone
These days my doctors have prescribed me opicapone in combination with
levodopa
. I currently take 600mg of Sinemet, 10mg of selegiline and 3 grams of Vitamin B1 per day. I wanted to ask who takes opicapone their experience in this regard.

“The three amino acids that are most effective at increasing dopamine levels are l-tyrosine, L-dopa, and phenylalanine.”
Anyone using these in combination? I think adding SAM-e to this might work synergistically? I’m learning other means of increasing dopamine to wean off of Azilect.

Hello, now I know I'm pretty new to the group as my wife was diagnosed on 12th March but given a PSP diagnosis from 29th Jan but she's had the symptoms for a while now and not seen a Neurologist due to Covid (blah blah blah). But she's been on Co-careldopa and that didn't seem to help, she's now on Pregabilin

Regarding levodopa/mucuna: is this true?
If
levodopa
is used as a lifetime remedy, your body's intricate system for monitoring hormones will be short circuited. Sensitivity of the monitoring system is degraded and the system itself is eventually disabled. [i]

NURR1: Facilitating the transition of neural stem cells into functional dopaminergic neurons.
//////What is NURR1? Nuclear receptor related 1 protein (Nurr1) is an important transcription factor required for differentiation and maintenance of midbrain dopaminergic (DA) neurons. Since decrease in Nurr1 function either due to diminished expression or rare mutation is associated with Parkinson's

Onset/diagnosis after years
Treatment with
levodopa
had short-lived effects so doctors decided to use DBS therapy. DBS did not have the desired effect; my aunt has electrodes in her brain that stimulate dopamine production.

Grapefruit reduced carbidopa-levodopa? Please UPDATE
Still relatively new on HU, I’m also searching through the old posts looking for positive experiences to optimize C/L efficacy. In doing so, I came across the multiple posts from 3 years ago about the beneficial effects of grapefruit juice. In particular, wriga's scientific findings and experiment seemed

two unrelated topics - Viraball & Mucana/Rasagiline
Doesn’t make sense to me that it would be ok to still be on Rasagiline if taking synthetic
levodopa
with C/L, but not on a natural form with Mucana. My doctor won’t engage in any conversations regarding Mucana. Any thoughts from someone that paid more attention in high school/college chemistry?

Journey of Confusion
My wife's journey of confusion started on the 29th Jan 2023 when FINALLY she was diagnosed with Atypical Parkinsonism - Likely Progressive Supranuclear Palsy, this was the day our world ended at 51yrs old........ No age I'm led to believe. We had been waiting to see a Neurologist since 2021 when we

PS128 yoghurt "recipe"?
He has been taking probiotics for the last year, and taking B1 +
levodopa
. He has a slow progression, and his still training for a half marathon. I don't know what is helping or not, but I won't take any chance and stop PS 128 because of the price.
