Clinical trial update

Trial Drugs: Inavolisib and Giredestrant. Today was my second set of scans (every 8 weeks) and the results of the chest CT and Bone scans were great, nothing new, all stable and NED in lungs. The abdomen CT showed slight increase in the size of my liver lesions. I am ER+, HER2 low +1, PR-, AR+, with pik3ca and TP53 mutations. This trial addresses the Pik3ca and ER+ and is my second line of treatment after Ibrance and Letrozole for almost 2 years. Can you guys share what 2nd line / 3rd line treatments have worked for you with similar subtypes? I am going to Cleveland Clinic at the end of the month for evaluation for the histotripsy procedure. But I will likely also be changing treatment soon as well. --Trish

I am on TRUQAP. Came out in November 2023. I have pic3 and another mutation. On 160. The 200 made me very sick. 24/7 diarrhea. Check it out. Reenie

Dear Rbeth---I'm still on heavy duty chemo because my bloods are not yet to where the oncologist wants them to be. So, it's more chemo instead of starting Truqap. I am so interested in how the Truqap is working for you. It's too bad that the doctor started you on a high dosage. How much do you weigh? I weigh 94lbs and so the doc sill be starting me on 100 mg rather than the full dosage of 200mg.

Jersey jazz- I am so sooty to hear you are down to 94 lbs. so tiny! I hope you feel well enough to put on so weight soon.

Dear friend Bettybuckets-----I never read/saw/heard the expression "sooty". but gather it is a bad thing. I have shrunk two and a half inches with a curvature of the spine, down to five feet and holding, hopefully. Two years ago, I lost ten pounds gradually, from our monster illness. Then, last year with the horrid surgical error, I lost twenty pounds in two weeks. Recently, with the latest liver crisis, I seem to be going downhill. However, I fully intend to regain some of my weight back and in June to get back into my beautiful motorhome with a friend from Scotland. Then, also in June, the QM2 will sail us back to the UK for an eighties celebration. (Not mine.). My eighty seventh birthday went beautifully with all able bodied family present for a sit down dinner in house.I somehow squeezed twenty -fourpeople into my dining room. That was no mean feat. Of course, there seems to be one person in every family that brings negativity to the occasion and this was no exception but I ignored her and kept the smile on my face. It really was a wonderful occasion.

Wowzer! That means I am really really impressed! Sooty was meant be “sorry to hear” my darn fingers! My son was just exploring Scotland and the pics looked so gorgeous- I bet you can’t wait to get back there in that motor home of yours! Can’t believe you had all that family over and in your dining room… reminds me I must make calls to set up my mom’s 90th in just 2 months! Thank you!

Wonderful that you mom is getting to be ninety. Now that I accomplished eighty-seven, my aim and goal in life is to achieve ninety.

The motor home will not be going with me on the QMary2 to Britain but will be aiming for the Florida Keys. Right after that, God willing, I and my friend will sail on the QMary2.

You are so right about the beauty of Scotland. It really is remarkable. Ireland has the PR people and the Scots just get on with it. It reminds me of Vermont and New Hampshire. Vermont has the PR and New Hamshire mirrors Vermont in beauty but is quiet about it.

Keep well, my friend!

The Histotripsy treatment looks very promising. Destroying tumors with sound is an exciting new idea. I remember learning about it here two years ago and have been interested in it since then. Hope it goes very well for you. Please share any updates on the Histotripsy treatment if you are able. I don’t have your cancer subtype, but I am having great success on the new ADC Trodelvy (Sacituzamab Govitecan) as a third line, Best wishes for a good outcome at Cleveland Clinic.

hi Trissh, it’s so interesting to hear about your trial drugs and the histotripsy procedure, none of which I’d heard about before, thanks for sharing!

I don’t have the exact same subtypes (am ER/PR+, Her2 low with Pik3) but in case it’s useful to hear, after starting on Ibrance (18 months) I went to Verzenio (3 months), Xeloda (6 months), and Piqray (5 months), Enhertu (2 months), now on Trodelvy - hope this one lasts a good while!

Wow it is amazing to see six new MBC treatments, which have only become available in the last few years or so, discussed in this post alone! Between all the new SERDs, ADCs and other treatments there is so much to celebrate and every reason to remain hopeful. 🍾🥂
