I believe this is the completion of the 2nd trial for the alternative dosing (5 on/2 off) of ibrance, just released TODAY! Someone who knows how to read these things, please tell me what I did wrong that I couldn't find their "conclusion" or "recommendation" after completing the study. Thanks for any light you can shed on this! I have trouble understanding all the jargon but for what I did read, it sounded very promising! Clarify please! <3 God bless you and heal us all in Jesus name, amen!

I’m on a 3 week on and 2 weeks off which is working well for me. I agree with you that I don’t see a conclusion to the study other than the side effects chart. The one thing I would not want to deal with is the Goserelin shot once a month. I’m not sure what Goserelin is, but adding another shot or appointment does not appeal to me. Maybe someone else can tell us more about this.

thanks for the response! I'm just wondering if NOT having the 7 days off is an issue. I'm thinking of being my own study LOL 5/2, 5/2, 5/2, 5/7 . This malarky will make you crazy! <3 xo

this is what I’m getting from previous evaluations : TAKE-HOME MESSAGEThis phase II trial investigating an alternative dosing schedule of palbociclib in patients with HR-positive/HER2-negative advanced/metastatic breast cancer showed that a 5-days-on/2-days-off weekly schedule reduced the incidence of grade 3 and above neutropenia and improved the tolerability of palbociclib compared with historical data of palbociclib trials. Serial monitoring of serum thymidine kinase 1 (sTK1) activity showed a significant reduction of sTK1 at cycle 1 day 15 and cycle 2 day 1, indicating that this alternative dosing schedule achieved effective target inhibition. The 5-days-on/2-days-off weekly dosing schedule of palbociclib reduced hematological toxicity without compromising efficacy in this patient population.

All I can say that this 5/2 schedule is working well for me. I’m not getting any other shots of fulvestrant or goserelin. I have my sanity, I’m comfortable, and moving forward. Recent scans were stable, and despite a variety of vaccines in the fall, my tumor markers are all within my best numbers.

I do not miss the neutropenia that was undermining my enjoyment of life. 🥰
