My take away is Nestlé is applying for a patent for a formulation of adenosylcobalamin to treat mitochondrial dysfunction and symptoms related to adenosylcobalamin deficiency which plays a role in the Krebs cycle ( energy production) . It sound like they are developing a probiotic that will make adenosylcobalamin in the body.

I have found it difficult to find studies and information about isolated adenosylcobalamin so I found this very interesting.

Many studies are summarized in the patent application that compare methylcobalamin effect to adenosylcobalamin.

I read someone's post on this form about adenosylcobalamin which is what prompted my research. Many of my PA symptoms are gone but I'm still left with exhaustion that comes on like a freight train but then goes away for a while, along with some other symptoms which may or may not be related. Could be hashimoto or perimenopause, not sure. Working with my doc but all other blood work looks good. My worst symptoms of PA have resolved.

I've only received methylcobalamin injections. I believe I've had life long PA but only got dx 2 years ago. Makes me believe I've likely never absorbed adenosylcobalamin in my life which may explain the extreme exhaustion. Or like I said, could be something else. I'm no doc but makes sense. Nice to find some info on the topic.

Adding adeno sublingual to my hydroxy injections has helped me a lot.

Can you share specifically what adeno has helped with that injections alone didnt?

I just started sublingual adeno yesterday. Made me feel really high for an hour or two and I didn't like it. Reminded me of the first and only b12 shot I ever got when I was 20 yrs old. Made me feel so bad that I never got another shot until 40 sum years old. If only I knew back then it's what I needed. I'll continue the adeno with high hopes.

It really lifted my energy as that was the thing that hadn't improved.

I thought we all had ( or those of us with good gut microbiomes) the bacteria to make b12. Problem is they are in the colon & we are unable to absorb it there.

I had done some research when switching from cyano to methylcobalamin and that research suggested that when using methylcobalamin, one had to supplement with adeno sublingual. Also gently sustain b6 but not too much. It all got to be too much to monitor so I switched to hydroxocobalamin because simply put, the body can convert it into the methyl and adeno it needs. After trying all three, I agree with Sally Pacholok that hydroxycobalamin is the best form of b-12 supplementation. I am older now but feel like my body is operating more normally.

Absorbing or ingesting adenosylcobalamin is not necessary. However, it is essential for human life to be able to produce adenosylcobalamin from the base cobalamin molecule.

If you ever became fully unable to produce adenosylcobalamin in this way then death would be virtually certain unless injections of adenosylcobalamin were initiated and sustained.

There are some rare genetic metabolic issues that can affect cobalamin conversion and transport. They usually present in infancy but rarely can have an adult onset.

But to put it more shortly : it's probably not something you need to worry about.

Hi, I’m confused by your response here. If you can’t absorb B12 - cobalamin - then you need injections. Hydroxylcobalamin converts to both methyl and adenosyl. If you only take methylcobalamin then you are not getting adenosyl. Surely you must then supplement adenosyl as vital for mitochondrial function?

Any B12, before being used in cells, is first broken down to the base cobalamin molecule and then rebuilt as adenosylcobalamin or methylcobalamin, as required. So no matter what form of B12 you absorb or inject, the body will convert it into the required form.

Is that because methyl can be converted to adeno? I read someone say that but haven't seen it in my research. I've had 1 cyano and 1 hydroxo IM injection in my life and also used sublingual b complex for a few months here and there in my life
