Olaparib hasn’t worked for me . Anyone had this happen and had success with another PARP?

Hi everyone,

I’m back of Chemo for 3rd line, this time with Carbo and Caelyx. This is just a year after 1st recurrence, Chemo reduced cancer to nearly NED, 7 months of Olaparib and then CT scan showed new tumour on spleen. CA125 36.

Oncologist mentioned later trying another PARP possibly Rucaparib. Anyone had success when one didn’t work ? Alternatively Tamoxifen was mentioned, should this work as well?

I would love to hear of success from anyone who had a successful 2nd maintenance when their first failed 😞

Hi Towergirl

In general in UK you only get one go at Parp inhibitors on NHS. Olaparib also failed for me.

The only way to continue to access Parps was to go on a trial. I did this in January 22 at the Marsden. Unfortunately, although Olaparib and the trial drug did work on the cancer they also gave me lung problems. So now on third line chemo. When I finish I will be looking for another Parp trial xx

Hi Lundy,

Thank you for your message and information. I did understood that I could not go back on Olaparib but not all PARP. Maybe my oncologist was thinking of a trail, something I must ask when I see him again. Good to have this knowledge thanks.

Hi Lyndy,

I was for 7 months on Zejula and I got pleural infusion on my left lung. Stoped Zejula. Since September I have no therapy. Now I am stage 4 ovarian cancer. Oncologist waiting for CA to increase/ now 24 / to started second line of chemo. I am wondering how can he live me waiting.. and he told me my life expectancy is months to one year. I don’t now what to do. ..

Morning Towergirl

I was on Niraparib for a year and it stopped working and I went back on carbo /paclitaxel. Diagnosed stage 4a in Aug 21 now spread to spleen and liver. My understanding is you can’t get another parp on NHS unless via a trial. I’m on the look out for one however my oncologist is proposing weekly paclitaxel for three week with a break on the fourth. Good luck xxxx

Hi Skyeplus,

Thank you for replying, my recurrence last year was also to Liver and Spleen . I was offered a 10 hr operation to remove spleen with liver scrap. Declined as not confident and didn’t want to lose spleen. I had Chemo and thought Olaparib would be the best way! Now I wished I had been brave!

Have you been offered an operation?

I’m having Carbo and Caelyx 1 cycle every 4 weeks for 6 months. Hopefully we both find our different chemo’s both removes all signs of cancer, Good luck for the future xx

I was on Niraparib for 3.5 years and then got my 2nd recurrence. Had my spleen and tail of pancreas removed September 2022, followed by 4 chemos. Oncologist said I can’t go back on parp ‘at this time’ but CA125 is holding at 10 🤞🏻

I have to take a low dose antibiotic twice a day but otherwise seem to be ok x
