Has someone of you dear MPN people been a part of a Navitoclax study? It is offered to me and I think I'm going to start with the study soon. They are taking me off Pegasyis and HU completely and starting with Navitoclax alone, without the JAK2 inhibitor. I'm a little concerned, but on the other hand, if I don't take this offer now, maybe I, 'll miss a good outcome...

Navitoclax
There are some posts that discuss Navitoclax and its related agents. Has Dr told you why this therapy is proposed? Has Dr said why not the better known combo with Rux?
On the other hand if they think Nav alone can benefit you, skipping the Rux is not entirely a negative.
One overall message is its potential benefit for at-risk non-driver mutations. Do you have mutations other than Jak2 with your PV? One mutation possibly relevant to Navitoclax is ASXL1.
Are you not responding to the PEG and HU?

These studies are usually with Rux combos, I've not seen as much about Nav alone:

"we provide preclinical evidence showing that the combination of ruxolitinib and ABT-737 (equivalent to Navitoclax) is a promising therapeutic strategy for MPN patients with concurrent JAK2V617F and ASXL1 mutations"
Hello EPGuy,Thank you very much for the answer and the useful links.
They suggested a study because it appears that my PV is progressing to MF with stage 1 fibrosis and an 18cm spleen. As far as I know, I only have the JAK2 mutation.
Navitoclax study, there are parts of the study for those already on RUX. Since I'm not, only NAV without RUX will be tried. We hope to reduce the spleen and fibrosis.
Currently on PEG, I have elevated body temperatures every day, a feeling of heat and burning on my skin, I can solve this with Paracetamol, but taking it every day is not a solution, because it creates an addiction. As far as I know I can always switch to RUX which would reduce my symptoms, but let's try NAV first and see what happens.
I’d go for it . They will closely monitor you. Good luck
