Progesterone - twice a day 25ml or 50ml at once ?

Hi ladies. My first IVF (successful) in 2017 I had Progesterone infection 25ml in the morning and 25 ml in the evening.

Now my doctor (new one) prescribed 50ml ones a day but he said if I feel comfortable I can do as before twice 25ml

What is your doze and what do you think is the best to split or to do one shot a day?

It would make to real difference if you took them together or just as 1 dose as long as you are taking them as directed by your doctor - 50mg dailyGood luck

Natural progesterone has two spikes: morning and evening. So if I were in your shoes and you can take the shots (they are more painful than stims, in my experience), I'd do it twice a day. Just to mimic the progesterone naturally works.

In my case, I only got 25mg once a day. I don't know why I got such a low dose. My cycle wasn't successful, so I wouldn't recommend. Do you know why you got a higher dose? Is that standard for your clinic?

Thanks. All injections I'm doing myself. So I get used to it:) to be honest I didn't ask why 50ml a day, I just followed but second round now I know which medication and doze work for me. So I'm trying to follow but wonder why doctor said one or twice a day doesn't matter. Maybe in the begging they prescribed 50ml cos I have PCOS?

I am taking 25mg 12 hourly, apparently the subcut peaks within 2 hours and starts to reduce after 12 hours as stated below. So by taking it 12 hourly you create a steady state. I hope that helps

Pharmacokinetic properties

Absorption

Progesterone serum concentrations increased following the subcutaneous (s.c.) administration of 25 mg of Lubion to 12 healthy post-menopausal females. By one hour post-administration of a single s.c. dose the mean Cmax was 50.7±16.3 ng/ml. The progesterone serum concentration decreased following a mono-exponential decay, and by twelve hours post-administration the average concentration was 6.6±1.6 ng/ml. The minimum serum concentration, 1.4±0.5 ng/ml, was reached at the 96-hour time-point. Pharmacokinetic analysis demonstrated linearity of the three s.c. doses tested (25 mg, 50 mg and 100 mg).

Following multiple dosing of 25 mg/daily via subcutaneous administration, steady state concentrations were attained within approximately 2 days of treatment with Lubion. Trough values of 4.8 ± 1.1 ng/mL were observed with AUCs of 346.9 ± 41.9 ng*hr/mL on Day 11.

I was prescribed 25mg injection every morning, but then supplemented by a 400mg pessary in the evening … 🤷🏼‍♀️ x

Thanks. For me after injection in the morning and evening I have pessary 200 mg
