Concentration of levodopa in plasma vs time curves for Madopar 100/25 IR, Madopar 100/25 CR, and Madopar 50/12.5 dispersible

Hello,

I have been trying (unsuccessfully) to find the above graphs. In case someone has them, I would be really grateful if they could share the data. A link to a peer-reviewed paper would be best.

Here is plasma vs time for Madopar 100/25 IR - the graph designated by the triangles. Placebo in this case refers to a medication added to the Madopar.

Keep in mind there is also an unknown delay to get through the BBB and for the levodopa to be converted to dopamine. Then there is an additional interval for that dopamine to fade away.

The blue one corresponds to the link above, full text available for download from Researchgate.net, free of charge. Thank you for providing both links.

Both graphs are not that dissimilar, I do not like how spiky they are, so it is difficult to decide whether to go through the ordeal of switching medications.

I will have to do the same comparison for Madopar plus Entacapone, hopefully that will help me decide.

Just noticed the missing values along the Y axis. The values are: 0, 100, 200 ... 900 micro gram per ml litre. (Software issue, probably).

The error bars on the C/L curve are pretty big and likely overlap the Madopar curve. In other words the differences between these two curves may be due to random variation. So not worth the trouble switching from one to the other.

The Flexilev system are doses dispensed in increments of 5 mg of levodopa, 1.25 mg of carbidopa, at programmed intervals. Multiples of this increment can be dispensed at once, Said to be approved in the UK but do not think it is being marketed there yet. Links:

Here's an app which draws the pharmacokinetic graphs for levodopa and a wide range of other drugs. For each dose, you enter the size of the dose and the time it is taken.

Hello,

Thank you for the link. I am aware of your App, I think it is a very good tool for calculating Pharmacokinetic graphs. I am surprised that the Neurologists are not required to use something like it instead of giving the patient a prescription and telling them that they know their body best, so they can dose as they see fit. This happened to me, and was the reason to develop interest in Pharmacokinetic calculations.

I am interested in using real data as opposed to simulated ones, and this was the reason for creating my post. I am particularly interested in finding the graph for Madopar dispersible which I have not been able to do so far.

I don’t know about Modopar, but I have been using the chart for Rytary and I have seen really good awareness and results. It also allows me to design my day and be more flexible in my routine.
