Hi All, Ron does not do well on Sinemet, so I've been substituting MP. His Sinemet is 25/100 and I just bought Now Dopa Mucuna and was using Solaray, but just guessing at dosage. But I have seen posts by several of you guys for formulas, but can't locate them now! He doesn't have much movement disorder, his is mostly cognitive, sadly been diagnosed with LBD in March.

Thanks so much, Hidden ! This is all just so daunting! Ron has had ADHD and OCD most of his life, so I also want to talk to his PCP, who has known him for many years. Hopefully, he can prescribe Something to address those, in tandem with his very by-the-book Neurologist. Any mention of these issues have fallen on his Neuro's deaf ears!
When buying your Mucuna Prurians/Velvet Bean 'standardized extract', you need to pay less attention to the total amount of 'seed extract' and more attention to the actual yield (%) of 'L-Dopa' (the critical ingredient). The Now product sounds ok at first glance - until you realize that you're required to take TWO capsules to get an L-Dopa yield of only 120mg (or 60mg per capsule). This is OK if you don't mind taking more capsules than necessary, but I try to streamline my already vast number of supplements and there are other brands that provide a higher yield (avoid any that don't include the l-dopa % info). I don't know if you're in Amazon territory but I recently found a product by 'Ayu Nutrients' that provides seed extract 600mg, 30% yield = 180mg L-Dopa per capsule (which I take 3x p/day). For LBD you should ensure your hubby's taking plenty of omega 3, and consider regular MCT/coconut oil (in coffee, smoothie, or food), and a good curcumin supplement (Longvida or Bioperine).
I've been reading old posts and found this eye opener from a Harleybob08 post 9 months ago. I'll have to study this for several days, but it raises serious issue with how we determine dosing and the need to take/balance seratonin when taking L-Dopa.
Do you know the ratio, i.e., how much 5-HTP should be taken with 100% pure L-Dopa?
I bought 50 g of 100% pure, pharmaceutical grade L-Dopa (and 25 g of 5-HTP from Nutrivita,) but as I read more, I find many pwp feel taking Mucuna Prueiens is preferable to taking pure L-Dopa, but I am hard pressed to throw both packs out.
I'm not aware of any particular interactions - positive or negative - between L-Dopa and 5-htp. I have no medical credentials but, as mentioned to Lmji who has a similarly pure L-Dopa product (below), I would start with a 1-to-1 comparative dosing with the 50-100mg typically prescribed for early-stage PD. Unless you have your own lab and are prepared to set up weeks/months of elaborate testing protocols, I'd recommend a simple process of trial and error (using the manufacturer's recommended dosage as a starting point) in order to determine your "optimal therapeutic range".
The researcher's (M.Hinz) focus on the "competitive inhibition state" (in this case how it interacts with other "amino acid intermediates"), is most likely driven by the fact that he has his own line of supplements to sell and tends to design his studies to result in greater sales of his product line. Along with my Mucuna, I include a common, readily available serotonin precursor in my supplement regimen (5-htp) and tweek as necessary...
Sorry Lmji, just saw your post. If you've got an extract that yields 98% L-Dopa, that's about as pure as it gets and should be compared 1 to 1 with the analog/prescription (Sinemet c/l) version to gauge the dosage that will work best for you. For example, someone recently diagnosed is typically prescribed Sinemet 25/100mg (Carbidopa/Levodopa respectively) and advised to start with half a tab (50mg of levodopa). So you should use 50mg of your 98% L-Dopa for starters in such a case and gradually increase until you find your ideal (50mg is about half the amount found in a medium-sized vitamin capsule). Consider adding a 100mg capsule of 5-htp to the mix 1-2x p/day. *BTW: I see you sourced this in China... via Alibaba?
A recent study's results: "When compared to LD+DDCI [dispersed pharmaceutical L-Dopa/benserazide combo], MP-Ld [Mucuna Pruriens low-dose] showed similar motor response with fewer dyskinesias and AEs [adverse events], while MP-Hd [MP high dose] induced greater motor improvement at 90 and 180 minutes, longer ON duration, and fewer dyskinesias.
