When do people tend to take their two days rest? between ending one week and starting another, or 'mid' week? Or, do people just tend to take one days rest between every run thus effectively completing each week in six days?

Do whatever suits your other commitments. I ran every second day - did not follow calendar weeks, but many others ran 3 days a week and followed the 'Weeks' in the programme, taking the weekend off.
I ran every other day until week 6, when I found I needed an extra day off between runs for a while, then I went back to every other day. The key thing is to listen to your body and do what suits you. Run alternate days if you want to (once I'd started c25k I didn't want to have more than one day off between runs!) but be preparaed to take longer rests when your body is tired. Best of luck.
Just go with your life diary, if you need a day off running well take it. It won't do any harm to have an extra day if a busy schedule gets in the way. Enjoy the programme.
As others have said, go with what suits you. As long as you're taking a rest day between runs, it's all good. I started following the weeks and taking two rest days before realising that I was getting tetchy on the second rest day and wanted to run, so from week 3 onwards, I ran every other day for the most part with taking an extra rest day when I needed to.
Thank you everyone. I don't really feel the need for 2 days rest at the moment, so I will try to push on at a run every other day, at least until the runs get longer! Having said that, I have had two days rest imposed on me this week due to the appalling weather and am starting to feel really frustrated. Seriously considering buying a treadmill!!!
