It takes 6-8 weeks for Levothyroxine to be fully absorbed and start working on symptoms. It's usual to have a thyroid blood test to check levels 6-8 weeks after starting Levothyroxine in case a dose adjustment is required. You should leave 24 hours between last dose and blood draw.
Dose adjustments are usually in 25mcg increments every 6-8 weeks when required. If you want advice on blood results post them with the lab ref ranges (figures in brackets after results) and post them in a new question.
Levothyroxine should be taken with water on an empty stomach for maximum absorption, 1 hour before food and drink or 2 hours after, 2 hours away from other medication and supplements and 4 hours away from iron, calcium, vitamin D and oestrogen.

As early as possible, I usually get to the hospital phlebotomy department for 08:00. Delay taking your dose at 07:00, and take it straight after the blood draw.
On this one occasion, don't worry about taking your levothyroxine shortly after food, etc.
