Severe joint pain while on Zytiga + dexamethasone + temazepam (for insomnia from dexamethasone) along with xofigo and xgeva
After taxotere, then 1-1/2 years doing very well on
Lynparza
+ Zytiga + Prednisone + Xgeva, PSA went up (last July - 5 months ago) so stopped Oliparib and switched steroids to dexamethasone (higher, 4mg. doses at first, now tapered down to 1mg) and added Xofigo for widespread bone mets.

Approved for Lynparza (Olaparib)
Sartor has me going on
Lynparza
probably starting at my next appointment in two weeks. If anyone has any advice as to side effects, time to take it, etc or any other advice, I would appreciate it. I know I used y'all's advice when on chemo and it helped quite a bit. Thanks

Lynparza? Mixed blessing.
After 9 months on
lynparza
for Brca 1 pc, developed moderate anemia. Scan shows reduction in bony Mets, which is good, but now have Mets in liver. Not good. Still have no pain. Ten years out and still have not had chemo. I guess that is next step. Anyone else with liver/?

Is anyone else having trouble getting your
Lynparza
? The AZ and me telephone refill line is busy with at least a two hour wait and once they answer they say they are sending the meds, but I still have not received them and going on my 3rd day without. I’m getting really nervous about it.

Lynparza (olaparib) slows progression in men with BRCA2, BRCA1, and ATM gene mutations
It was an interim analysis, but results were especially good for men with BRCA1/2 and ATM mutations. A similar analysis showed the benefits on BRCA 1/2 of another PARP inhibitor, Zejula (niraparib): https://pcnrv.blogspot.com/2019/10/olaparib-slows-progression-in-men-with.html

Antitumor Activity Seen With Keytruda in mCRPC
inhibitor-based combination regimens have shown activity in mCRPC, including the CheckMate 650 trial of nivolumab (Opdivo) plus the CTLA-4 checkpoint inhibitor ipilimumab (Yervoy) and KEYNOTE-365, which showed promising antitumor activity and well-tolerated TRAEs when pembrolizumab was combined with olaparib (

Next line of treatment: too many options
Option 3: Try and be a part of clinical trial 16U05 (if biopsy shows BRCA mutation) (https://clinicaltrials.gov/ct2/show/NCT03012321) which, depending on which arm he is placed in, could mean being treated with only olaparib (
Lynparza
), or only Zytiga, or both
Lynparza
and Zytiga at the same time.

Approved Treatments for Triple Negative MBC Patients
Patients with inherited (“germline”) BRCA mutations may want to speak with their medical teams about taking a PARP inhibitor such as Talazoparib (Talzenna) or Olaparib (
Lynparza
), which are FDA-approved for HER2 negative MBC patients with BRCA mutations.
