What is 50% person in PMR/Pred terms please?

Hi Polly-peeps...

When I spoke about my high starting dose of 40mg, to get relief from pain, in a previous post, someone made the observation that I may be a 50% person?

I was wondering if anyone could elaborate on this? Or point me in the right direction to read about it?

Many thanks 🙏🏻

The more knowledgable among us will be along soon. But meanwhile as you have not had a reply since posting two hours ago I will give it a go! From what I’m understand we are all different in how much prednisolone we can absorb. If you absorb it all you get the advantages of the full dose eg15mg. But if you are a 50% absorptionperson youwould only have7.5 mg helping you along... not enough in the early days. Hope this helps and good luck.

Mary.

PMR since Nov 2015. Diagnosed Jan 2016. Now reducing DSNS from 8.5 mg to 8mg

One of the pharmacodynamics (the branch of pharmacology concerned with the effects of drugs and the mechanism of their action) factors that is important in how anyone responds to a drug is how much of the dose being taken you actually absorb. In the case of pred this can vary from 50% to 90% - so one person taking a 10mg dose may get the benefit of 9mg while another may only get the benefit of 5mg. Which obviously makes a big difference to the effect they experience.

If it was me I'm surprised I didn't explain - even if it was earlier in the thread.

Thanks very much... there were a lot of points in my rather long post, and I appreciate your original informative post 👍

It’s such a steep learning curve at first..🤔

Mary has already explained it well. I see that it was PMRpro who raised the possibility of you being a "50% person" in her reply to your original post, and I'm sure she will be along shortly to explain/elaborate further.
