When my muscle weakness snd neuropathy were getting worse I injected more with no relief.... it was my folate!...Do not divgolate acid it needs to be mythyl folate. Also check for high B6 levels and low electrolytes... surprisingly these have same symptoms as PA and are sffected by PA which is why it is so maddening to solve

Hampton77, glad that you found a solution that worked for you but please bear in mind that in general folic acid is as good as methylated forms and some people don't respond well to methylated forms.
There are a huge number of genes that control steps in the processes that go on in your cells. There are also a large number of variants on those genes which means it is impossible to generalise about which form of a particular vitamin is 'best' because it will vary considerably from one inidividual to another.
The genetics of B12 processing are strictly off topic for this forum, as it is about B12 absorption problems. Even more so for other vitamins.
It is possible that that genetic variants may be involved in some of the problems individuals have post loading shots with maintenance regimes but research into these areas are in extremely early stages and the general conclusion from studies tends to be that more research is needed to understand the mechanisms and processes involved.
Problems with B6 overdosing are well known and referenced periodically on the forum.
