Recent PGK1 and Terazosin Study

PGK1 and Terazosin has been discussed before on this site. A new study (link below) published on Aug 21st, 2024

The study, published Aug. 21 in Science Advances, presented evidence that PGK1 is a “rate-limiting” enzyme in energy production in the output-signaling branches, or axons, of the dopamine neurons that are affected in Parkinson’s disease. This means that even a modest boost to PGK1 activity can have an outsized effect at restoring the neuronal energy supply in low-fuel conditions—and the researchers showed that this could prevent the axon dysfunction and degeneration normally seen in an animal model of Parkinson’s disease.

I may not be understanding this very well but does this article advocate for using Terazosin to help with Parkinson symptoms?

it suggests that stimulating PGK1 will help. It also suggests that Terazonin will do this. However this doesn’t go as far to say that it should be prescribed possibly because of potential side effects and because it is not FDA approved for PD.

Terazosin, doxazosin, and alfuzosin (Tz/Dz/Az) are α-1 adrenergic receptor antagonists that also bind to and activate a key adenosine triphosphate (ATP)–producing enzyme in glycolysis. It is hypothesized that the increase in energy availability in the brain may slow or prevent neurodegeneration, potentially by reducing the accumulation of alpha-synuclein. Recent work has suggested a potentially neuroprotective effect of the use of Tz/Dz/Az in Parkinson disease in both animal and human studies. We investigated the neuroprotective effects of Tz/Dz/Az in a closely related disease, dementia with Lewy bodies (DLB).

I read about Terazozin over 4 years ago and got the approval of the Neurologist to change Tamulosin which he had been taking, to Terazozin at our request.

As with anything with PD I’m hoping it slows progress. My HWP does a myriad of things to help. Hopefully they all play a part!

Just walked 6000 steps up hill from a restaurant in France. He actually started jogging! It’s so strange on the whole he is doing so well, still ongoing symptoms which he tries to ignore. Enjoying the wine here in France dx 9 + years and doing well thanks. Not perfect but ok 👍!

We got the recipe from Wriga and joined in his research group. My husband does still use it. We make it ourselves.

I had to stop taking terasozin due to dizziness when exercising.
