Has anyone taken Modafinil or Provigil for fatigue? And if so did it help or is there any side effects? Thanks for the info. Karen

Interesting:

en.wikipedia.org/wiki/Modaf...

" it was found to significantly affect only the dopamine transporter (DAT), acting as a dopamine reuptake inhibitor (DRI) ..., modafinil increases locomotor activity and extracellular dopamine concentrations in animals...and also inhibits methamphetamine-induced dopamine release... Another human PET imaging study similarly found that modafinil occupied the DAT and also determined that it significantly elevated extracellular levels of dopamine in the brain, including in the nucleus accumbens.[53]"

Park Bear, So what is your opinion ? My dr has proscribed it for me but I’m waiting until the school year ends to try it just in case there was a weird side effect or if it isn’t for me and I reacted differently to it. Hence my question, has anyone been given this for fatigue? K

I do not have personal experience with it and would defer to those who have. That said, it looks like a reasonable idea - per the quoted passage it slows down dopamine reuptake so it could be helpful for Parkinson's. We certainly need a good medication for Parkinson's fatigue. If you try it, do let us know your experience.

I did a bit more looking around. I would not call this a complete survey:

ncbi.nlm.nih.gov/pubmed/198...

The Modafinil group had a statistically significant improvement on ESS [Epworth Sleepiness Scale] (p < 0.05).

On the other hand this next study failed to reach statistical significance. There did seem to be a non-statistically significant improvement. Bear in mind that failure to reach stat sig does not prove that treatment does not work - what it says is the study failed to prove that the treatment does work:

jnnp.bmj.com/content/76/12/...

Results: Of a total of 40 subjects (29 men, mean (SD) age 64.8 (11.3) years), randomised to modafinil or placebo, 37 completed the study. Modafinil failed to significantly improve ES[Epworth Sleepiness Scale] scores compared with placebo (2.7 v 1.5 points improvement, respectively, p = 0.28). ... Adverse events were minimal.

PB, I started taking the Modafinil 1/2 pill in the morning . Supposed to take a whole pill in the morning but that’s me trying to have control taking 1/2 to start. I do believe it’s helping . I have been feeling strong not so worn down. A couple days I have tried taking the other half around 11:00, so taking a whole pill but broken into two times. This weekend I am doing a Spartan Race in Monterey with my son, so after this weekend I will try the whole pill in the morning and see how that does. I am on (according to the pharmacist) a very low dose. I’m excited about this so we will see. Karen
