I’m due to do R3W6 - the 25 min run. I had an unintentional 4 day rest period, now I’m apprehensive about being able to do it. Surely I won’t have lost all that fitness that quickly? All mind of matter I guess.

No you won't.
As stated in the guide to the plan, healthunlocked.com/couchto5... you lose no significant condition in the first two weeks of non running.
After that loss of condition is gradual.
You will be fine, you mind will try and say you cannot do it, but the plan has got you this far and will take you further.
your body will probably enjoyed the extended rest and be ready. Just remember your pacing, not too fast
