Good question, all I can offer is my experience. I didn't weigh my self during the programme because I wanted it to be about fitness rather that weight but I just graduated so got on the scales this weekend.
Before C25K I walked the dog around 6-7K every day, my routines now is to run on alternate days but still walk on non run days.
Before I started running, 6K walking would just about maintain my weight (if I watched what I ate as well) since I started C25K I've lost around 8lbs in 9 weeks. I'm, not dieting but running has made me more conscious of what I eat. I still eat crisps and bread, curry, occasional cake, chocolate and of course I drink wine- I just don't go mad (as a yo yo dieter all my life this is quite a surprise!) and I try not to do compensation eating.
I know some will not lose any weight at all but probably feel a lot sleeker. The body is very good at keeping hold of its fat supplies but running is definitely helping get rid of my middle age flab. I'm looking forward to my first non weight gain Christmas ever!
The best way to find out how your body responds to running is to do the programme. At around week 5 you'll be getting into some serious fat burn exercising but you should feel some benefits before that as well. Just try to avoid rewarding yourself with rubbish snacks.
You burn calories as you sleep, but you burn a lot more as you exercise.

I walk a lot. A 5.5 hour jaunt of about 30km burns 1600 Calories for 73kg bodyweight.
