A pretty simple explanation of the mechanism and how it may avoid BTK mutation causing tolerance.
Wonder if degraders will help with T Cell Exhaustion as well.

Thanks, a decent summary from Dr Danilov. As he explained, the effect of the degrader is to grab the entire BTK protein and chuck it in the "recycle bin" (shuttle it to the proteosome). The trial drug NX5948 showed no major toxicities through dose escalation, so seems to have few off-target effects. I don't see any reason to expect the same benefit to T cell function as seen with Ibrutinib. However, healthunlocked.com/user/gar... did note the immunomodulatory activity of another BTK degrader in development NX2127, and she would have a better perspective.
From a couple of months ago, healthunlocked.com/cllsuppo... gives a comprehensive review of what's happening around BTK degraders for CLL/ SLL.
