Understanding Acalabrutinib

I have been on ibrutinib for 2 years with the only side effect being frequent rashes of varying types, but they are annoying and make sleeping difficult when severe.

My understanding of Acalabrutinib is that it is a more recent form of ibrutinib in which various side effects have been eliminated or reduced. Does it have more curative properties, or lose any of the properties that ibrutinib currently contains?

Am I understanding this correctly?

Thanks - Dan

Here is a 2021 video interview presentation by Dr. Matthew Davids on Acalabrutinib vs Ibrutinib for CLL discussed and points highlighted on CLLSociety site:

Also, here is a discussion by our members regarding rashes with Acalabrutinb that I hope will be beneficial to you in regard to your rashes.

Many thanks, cllady01. That gave me more than I expected. I have printed it and copied the link to present to my oncologist at my meeting in a bit over a week. Of course, there will then be a battle to get the government to switch which one they pay for, although I believe the costs are similar. I am in del 17P so it is very applicable.Your help on this is much more than I could have expected. Again, many thanks.

Thanks for this as well. I've read a number of comments on rashes on this site - all good stuff.

Hi Dan,

As I covered in healthunlocked.com/cllsuppo...

CALQUENCE Met Primary Efficacy Endpoint in Head-to-Head Trial Against ibrutinib in CLL

"In this first head-to-head trial of BTKis in CLL, Aca demonstrated non-inferior PFS with less cardiotoxicity and fewer discontinuations due to AEs vs Ib"

In other words, acalabrutinib is as good at controlling CLL as ibrutinib, but with a lower adverse event profile, which importantly includes less risk of developing atrial fibrillation.

Thanks very much Neil. I haven't seen that information and very much appreciate it. Although I suspect we know a lot more about it among our CLL group I'll discuss it with the oncologist. I very much appreciate you knowledge on many questions you have answered to various of our members.Cheers - Dan

I was on Ibrutinib for 14 months with rashes as well as many other side effects. Almost all the side effects were gone the day I started Alacabrutinib, however, the rashes that were on both legs are still there. They don't bother me.

Thanks for the note. It sounds like it is worth looking at. My next step is to see if the medical system that is paying for my ibrutinib will allow a switch.
