pioglitazone and adrenomyeloneuropathy
I assume you're referring to the 2013 article "Pioglitazone halts axonal degeneration in a mouse model of X-linked adrenoleukodystrophy" academic.oup.com/brain/arti...
My understanding is that MIN-102 (aka leriglitazone) was developed as a more potent version of pioglitazone. As we know, the 2-year MIN-102 study produced marginal results, so it seems unlikely that pioglitazone would be a breakthrough.
Minoryx (Leriglitazone) has successfully completed a phase 1 clinical trial showing good safety, tolerability and CNS engagement of PPAR gamma receptors at levels equivalent to those required for efficacy in preclinical models. It has reduced cerebral lesions in young males and younger AMN males have shown improvement: minoryx.com/media/minoryx_p...
It's the advanced AMN that it has not. I have participated in the last phase and am now enrolled in the advanced trial to see if it can reduce my adult active cerebral lesions.
The brain penetrant PPARγ agonist leriglitazone restores multiple altered pathways in models of X-linked adrenoleukodystrophy
