I have brca2 mutation. Has anyone had this and been treated with lynpraza? How was your response?

Do you need to have progression of disease to be eligible for this drug?

Hello - My husband has been on Lynparza since Jan. 15th 2022. He's doing great. PSA has dropped!!! He has a mutation on the BRCA chain. No side effects so far. Hope to ride the Lynparza train as long as possible. The worst part of this drug is ordering it. GOOD LUCK!

240 to 78 to 43. After the first month MO started checking CBC, Liver and PSA every 2 weeks. Good luck to you!

Also...he's still on 2 pills in the morning and at in the evening. Sometimes MO will reduce dose if you are having an issue.

The drug is approved for cancers with HRR mutations (BRCA1, BRCA2 etc).

Cancers with BRCA mutations have the best response to this drug.

It is only approved for men with metastatic castration-resistant prostate cancer

(mCRPC) who have progressed following prior treatment with enzalutamide or abiraterone.

BRCA2. Put on Lynparza at time of initial diagnosis via genetic testing in Feb 2018. At that time the drug was not yet FDA approved, and was only available via clinical trial. My MO was able to get "off-label use" for me due to my mutation. Was on the drug for almost 2 years, most of which my PSA was undetectable, before it ran its course. A great response while it lasted.

I ve been an Lympreza for a little over two weeks and have have PSA tested today so I’ll update when I have results. I failed Keytruda after 10 sessions but lucky to have the new mutation.

Please please do- what happened to you psa after 2 weeks? Do you have BRCA2 as l do?I would really be interested in your progress.

I am BRCA-2 and did the Lynparza, initially I had great and dramatic results, PSA dropping to under 4 in less than a month, from somewhere in the 20's or 30's... I don't recall the precise moment on the roller coaster. Anyway, I started it on August 5, 2021 and it stopped working in November.

Also yes I had to have failed (shown progression) after two rounds of ADT before my insurance would pay for the drug. I was declined after one round of ADT, then we tried Xtandi which worked for less than a month, after which I qualified and got the drug.

Minimal side effects and it worked great while it worked for me.

Not sure why it stopped working, my PSA was down to 1 in early November. I went off the Lynparza on 2/1/22 and haven't been on any treatments until starting a new round of chemo three weeks ago. PSA now 108.

I started on Lynparza in mid February 2022. So far, so good. All of my other meds have failed so quickly. I hope this one last a while!

My husband is BRCA-2 and started on Lynparza August 2020. Prior to Lynparza he was on Aberaterone/Prednisone which failed after 4 months. Prior to that they tried Provenge. His PSA has never been higher than 22 but since the Lynparza the PSA has been undetectable. Additionally he had 6 Xofigo treatments from May 2021-October 2021. His latest scans also showed no new lesions or progression of the bone Mets. I hope that your treatment goes well and that you’ll continue to show improvement to undetectable as well.

Been on Lynparza since June 2021. PSA started at 16 and dropped over 6 months to 14. More importantly my scans showed no progression. I needed to stop Lynparza due to surgery to replace my hip and femur last month. In the month my PSA shot up to 40. I just started back on it and will let you know if it works. Side effects were minimal.
