Anyone taking Lynparza?

I care for a man with highly metastasized prostate cancer. He has recently started on Lynparza. Would like to speak to others on the medication. I’m trying to determine if the side effects get any better. It’s been a rough couple weeks.

It can have really bad side effects, but it does a really good job in men who have BRCA mutations. It's a difficult balance.

I have been two months on the drug. No, the side-effects have not gotten better.

Constant nausea for me despite strong anti-nausea meds. Generally listless.

Anemia is the most common side-effect and I am heading toward that. Getting harder to get up in the morning.

This is a toxic drug. As reported, a few others can handle it better. Not sure I want to continue with it, but my options are getting fewer and fewer.

He’s BRCA-2. He was on Cassadex for a year and a half, then Zytiga for 2 1/2 years. We tried three months in a clinical trial on Abiraterone to see if it could get the Zytiga working again. Slowed the progression but didn’t stop it. That’s when dr. did the gene test. But not sure the side effects are worth the limited time it could buy us. Leaning toward just switching over to Xtandi. He had a rough first couple months on Zytiga, but his body adjusted. But if he’s not going to adjust on this I don’t know if he can take it too long.His PSA started it over 1500. He got down to .03 on the Zytiga. We are back up to .68 right now. I know it’s still low but we’re seeing new lesions.

Consider trying bat (bipolar androgen therapy) and if that shows an effect, straight testosterone therapy.

Not a lot of docs will use these therapies. Docs who have worked on Denemead's bat trials will do bat. Sartor at Tulane will do straight testosterone therapy if appropriate.

Found to be BRCA2+, and was put on Lynparza as a result. 2 years. All manageable side effects.

I'm also BRCA2 and have been down same road. I went on Lynparza after Xtandi stopped working. It slowly did it's job lowering my PSA without side effects but only worked for 10 months. I am currently getting ready for UCLA trial for Opdivo and Yervoy praying that it will work for me. Unfortunately you don't have a lot of options and the way my oncologist put it, Zytiga and Xtandi are so similar that if one isn't working the other probably won't either.Good luck!

Thanks for the info, but his has metastasized. It’s in his skeletal system from his pelvis to his skull.

Hi Nalakrats:Looks like my Zytiga is failing. PSA went up for 2 months - 4 to 12 - and now leveled off at 12. Is the next stop Xtandi???

Thanks for all this information. I will research it. And just to clarify, it’s in his skull but not his brain. So far we’ve had no organ involvement. Just his bones and lymph nodes. The skull lesion is new. Otherwise his lesions have maintained or shrunk.

What is a crossover drug to go with the chemo? The interesting part about this is physically I feel fantastic lots of exercise lots of golf. We all have crazy journeys- mine now for 18 years. Always appreciate your thoughts.God bless you

I am BRCA1 and running out of options myself. I was feeling pretty good so I took a gamble that I could tolerate the side effects. Man was I wrong, I started early September and by mid October I had to quit. the side effects took me down. The worst of it was breathing problems, shortness of breath. Other issues were fatigue, lots of bruising, weight loss, loss of appetite and desire to eat, aches and pains all over. I'm sure I missed a few but that covers most of it. I am also heavily metastasized, mainly my spine and femur. I am going on Casodex and hope that will slow progression for a bit. My other option could be Carboplatin. Good luck to you, I hope it works for you and the side effects are minimal.

Scans this week then meet with MO. What do you mean High T treatment to resensitize the cancer??? I will ask him.
