Is anyone on the UCLA trial for Opdivo with Yervoy for PC?

I have been approved to start trial and was wondering who else out there has or is going through it and can share their experience.

One should talk very carefully with the doctors about the negative side effects of combining these drugs. Two checkpoints inhibitors are blocked by this combination which theoretically could facilitate the attack of normal tissues by the immune system.

Preliminary results showed modest efficacy and 40 to 50% of the patients had grade 3-4 side effects.

Historically, there was a Phase III clinical trial of just Yervoy (ipilimumab) vs. placebo for advanced prostate cancer in a larger population with results reported circa 2016. Efficacy and Overall Survival benefit was low, and reported side effects were pretty high. It did seem to hold some promise for a small subset of responding men. Per the above, these current trials are hoping to find if some combination of other agents might work better in selected subsets of prostate cancer men, whose tumors typically run "colder" than other more virulently growing cancer types.

(For Gee Whiz anecdotal information, I will be starting 4 rounds of this same combination for my own metastatic melanoma, which I've had for the last 1.5 years on top of my 7+ years of advance prostate cancer. Earlier this month I stopped Keytruda at round #14 after my scans showed very probable metastatic melanoma disease progression. I'm still chugging along after 3+ years on Xtandi for the prostate cancer, but my labs are showing that it's becoming less effective, too. As an "advanced" patient with two deadly cancers, I will be getting a similar treatment, ... just outside of this closely monitored and controlled clinical trial being offered at UCLA for prostate cancer. Are you familiar with the mayo clinic case study of a melanoma patient who declined all traditional oncology treatment at the mayo because he wanted to use IP6 exclusively? After three successful years of using IP6 doctors at the mayo published a case study about two years ago describing this patient’s experience. The doctors are now doing a clinical trial of IP6 with melanoma patients. Of course, IP6 is very controversial because it has been viewed as a naturapathic remedy and is available over the counter. No profit for big Pharma in a naturally sourced medicine. I have been using it since 2015 with my prostate cancer. See my posts in the past.

Thanks for the reply, CalBear74. Personally, I would want more solid information as to the efficacy of this supplement for any cancer in humans. My general inclination is that single anecdotal experiences seldom meet the necessary level of evidence for efficacy, even if eating a boat-load of the stuff is unlikely to poison anyone.

Most of the sellers of IP6 state that their reportedly active ingredient usually comes from rice bran. Sellers in the USA also cannot legally misrepresent their product, and therefore include: "These statements have not been evaluated by the Food and Drug Administration. This product is not intended to diagnose, treat, cure or prevent any disease."

If others wish to take it, it's a choice. My personal situation with both advanced metastatic melanoma and advanced metastatic prostate puts me in the realm of looking to treatments with more solid levels of evidence for efficacy and treatment power.

I’m currently in a clinic trial testing Opdivo alone in conjunction with Docetaxel. I’ve finished my chemo but continuing with Opdivo every four weeks for next 24 months. My dx in 3/2020 was Gleason 9, mHSPC. While my treatments are not a direct comparison, I can attest that so far I have had no side effects from Opdivo. I do have some minor neuropathy and major thrush that is attributed to the docetaxel. My hope is that these will clear up with time. Good luck with your trial.

a heartfelt "GOOD LUCK WISH" for positive results

as a p.s. -- I'm 5 years into a "at the time one off" in situ injection in my hemi-cryoablated right side GL10 tumor of the aforementioned 2 drugs plus Keytruda. Akin to an experimental pre-emptive metastasizing strike.
