Have question about drug Talazoparib.

Does anyone have any personal experience with taking Talazoparib alone? Or info or involvement in trial using Enzalutamide and Talazoparib?

It's only approved for breast cancer, so men with prostate cancer can only get it "off label" (very expensive) or in a clinical trial by itself (below) or randomized (to placebo or talazoparib) with enzalutamide (below).

If you want to talk to a man who has used it, you can ask the investigator of the Phase 2 trial if anyone has given consent to be contacted. It's a small trial, so it is unlikely,. On the larger randomized trial, no one knows for sure who has taken the drug.

Does your cancer have DNA repair defects such as BRCA, ATM, CHECK2 etc?

This type of drugs are mainly effective in cancers having this type of defects.

I ask, because personally, I will not enter a clinical trial with this kind of drugs unless the cancer has DNA repair mutations, due to the undesirable side effects and I believe a low chance of benefits if these defects are not present.

my dad has an ATM defect and did a trial with avelumab + talazoparib. it did not work for the ATM mutated patients and caused a lot of bad side effects which cleared almost immediately after stopping talazoparib. that’s my only real knowledge of it.

I am on the Talapro2 trial which is enzalutamide and talazoparib/placebo. As Tall Allen said dont know if I am on talazoparib or not. It's been 6 weeks and the only SE I notice is fatigue and a small bit of neuropathy. My PSA dropped for 14.7 to 1.1. I will have scans next week to compare against baseline prior to starting trial.

I commenced the same trial this week. I took the second day of tablets yesterday. Except for a little of fatigue I have no other side effects. I am looking forward to excellent results like you.
