Is anyone on or been on Ibrance at 75mgs. 5 days on/2 days off or one week on/one week off with any success? I would appreciate any information.
Hope you are all having a good day.

When I started my Ibrance, I personally chose my regimen. That is 75 mg one week on, one week off. Then, the next cycle was 2 weeks on, one week off. And then next cycle and all after have been 3 wks on, 1 off. I fully intended to repeat this acclimatization process at 100 mg and then 125 mg but my doc said that since I am responding well to this regimen, I should stay on it. So that is what I am doing and I have negligible side effects. I wish you luck!
Thanks so much for the reply. Great information and makes so much sense. We have done the opposite. Started at the highest dose with the longest time on. 3wks on 1 off. I had no side affects except for some slight hair loss (not noticeable) and the immune system was bottomed out.
We have, since February tried adjusting the dose down and waiting in between for my immune system to recover. (one or two weeks). So no consistency. Someone else wrote that they do 5days on/2days off and that works well.
One of the ladies on this forum mentioned this trial to me.
Pfizer and Washington University School of Medicine have conducted a trial of this dosage which is now finished but the results have not been posted as yet. The theory is it is more tolerable and with less frequent high grade neutropenia and is more consistent because there is not a week's break in therapy,
There is an explanation of the trial at ClinicalTrials.gov
I am excited about this because I wanted to stay on Ibrance if possible.
Also, my referral oncologist told me she has a patient on one wk. on/one week off that works very well.

I was always on 75mg due to my neutrophils. As you know I was in one week on one week off but it did not work for me. That would mostly be because of my neutrophils as they still dropped off and I would occasionally need two weeks off. Good luck.
Thanks Sarah. We didn't try that. My oncologist felt we should move up to Xeloda. I went for a second opinion and the oncologist there has a patient on your dosage and it works.
I hope my oncologist will let me try it. The other alternative is to try exemestane before switching up to Xeloda. So some options.

Are they switching you because of progression? If so then the week on week off may not be a good option but it’s nice you have some. Abraxane was the only option I was given.
Sorry I missed your question until now. Yes I have had progression in my left lung first noticed in August and then in October. The progression is slight. My cancer has been described as very indolent. I would love to try the 5/2. There was a trial at Washington University School of Medicine with Pfizer. The trial has ended but results have not been posted. The trial was proposed because a significant number of people required dose reduction and sometimes discontinuation. The 5/2 regimen was suggested because the dosage is basically the same over 28 days as 3/1. In addition they think the 5/2 schedule is more tolerable with less frequent high grade neutropenia and dose interruption/reduction.
A lot of posts here have had great success with also disease reduction. I haven't read any negatives about this.
I hope the results of the trial will be posted early in 2021 and I hope I can try it.

Hi June
After starting, and persevering with, the higher doses I eventually had to settle on the 75mg Ibrance dose because of low neutrophils. I have scrapped in most months with 3 weeks on/1 week off and once or twice with two weeks off. Besides the low neutrophils issue the major difference on the lower dose was the fatigue was far more bearable. I’m stable mable so happy with the lower dose. I hope you find a dose regime that works for you. Vicki
I started on 125 but had low neutrophils. I've been on 75 on a 5/2 schedule for about 18 months with good results (remission). Neutrophils are consistently low but within the standard range.
