Trials of CALR and Jak2 selective agents

There have been posts on both these INCYTE drugs, including INCB160058 for Jak2 posted by member Coffeeglass.

and INCA033989 for CALR in several posts. Both are seeking better targeting of the mutation.

What's new is the Jak2 trial is underway:

It's US only in a few locations. For members with MF with lost response to Rux, this could be worth inquiry.

This CALR drug is not the new CAR-T cell therapy category discussed in different threads.

INCYTE is also the Rux guys. Surprisingly INCA033989 (the CALR one) is apparently an oral monoclonal antibody. I thought mAb's, and biologics more broadly, were always injections like IFN, but a quick check finds that oral mAb's are indeed a new thing. INCB160058 is an inhibitor, well known in oral form, possibly a better refined take on Rux.

This plot is from the prior thread, in vitro only. But it shows a dramatic effect and that there is a best dose (too much INCB160058 and the blue healthy cells on the right become wiped out same as the mutation)

As I noted the most important advance may be the new ability to ID the mutation. Once it's uncloaked there may be various ways to go after it.
