I increased my dose to 300 mg daily.

I just started increasing my melatonin dose to 300 mg daily. I do take it throughout the day. I have 60 mg capsules with pure melatonin except for the capsule. I'll see if it improves my labs which will be in March.

My cholesterol labs and a1c. I used the Cleveland clinic heart labs. Very thorough. I'm trying to get my A1C under 5.

Thanks for posting about this. Seems like a high dose to me, but I'm open to high dose melatonin and I'm curious about your experience.

Are there any updates on how you are doing at 300 mg/day of melatonin? This is nearing the dose that Dr. Shallengerger gives to his stage 4 cancer patients of 360 mg/day. I can make two suggestions, one for the A1c and one for the cholesterol if you are interested. High melatonin levels when eating may be counterproductive to at least one of your goals and may cause glucose intolerance.

Yes, your other replies give an idea of why I said that high melatonin levels when eating may be a problem in terms of glucose tolerance.

You didn't answer how you are doing at 300 mg/day?

Everything's going good. I get my cholesterol labs and a1c labs on April 10th and results April 24th

If we should avoid eating and digesting food close to our evening endogenous melatonin rise, what are the implications for using exogenous melatonin as a sleep aid? There is evidence that taking melatonin also leads to impairment in glucose tolerance. Rubio-Sastre and colleagues12 studied 21 healthy women, giving them either melatonin 5 mg or a placebo in the morning and evening on different days. An oral glucose tolerance test monitoring glucose and insulin levels for 3 hours began 15 minutes after each dose. Finding that the melatonin use was associated with elevated glucose levels at both time points, they concluded that the optimum timing for melatonin doses should be at least 2 hours after the last meal.

The point being that eating while your melatonin level is high is likely to be a problem. Taking melatonin before bed and well after eating should not be a problem. Melatonin secretion from the pineal gland increases with darkness generally peaking around 1:00 am to 3:00 am. Taking melatonin just before bedtime should be in line with the pineal secretion of melatonin. I reserve daytime use of melatonin for specific issues.

Melatonin is a hormone produced by the pineal gland in the brain. It helps to regulate the internal clock of the human body which controls natural sleep/wake cycles. Melatonin also plays a role in glucose control (how the body processes sugar and controls blood sugar levels): it acutely reduces the ability of the body to take up sugar from the blood after a meal. Under normal circumstances, this has little effect because melatonin is only high at night, usually after their daily food intake. However, certain groups typically eat when melatonin concentrations are high, including shift workers, representing 10% of the work force, the millions of people who consume food after dinner, and people using melatonin supplements against sleeping problems. In these situations, the effect of melatonin on glucose control may lead to high blood sugar levels and increase the risk for type 2 diabetes (T2D). In fact, geneticists have recently found that people who have certain genetic variations in the gene MTNR1B (encoding melatonin type 2 receptor) have an increased likelihood of developing T2D. Thus, the proposed study is aimed to determine the interaction effects of melatonin and MTNR1B in glucose control and explore the underlying mechanisms. The results from this study may provide vital insight into the development of interventional strategies and personalized recommendations against the increased risk of T2D, especially in shift workers and later eaters.

Melatonin is not only made in the pineal gland and in fact the pineal gland is one of the lowest producers of melatonin. Melatonin is produced in the gut at a rate that is said to be approximately 400 times that of the pineal gland which eclipses pineal gland secretion by a tremendous amount. Melatonin is also produced in the mitochondria and this level is thought to eclipse the production of melatonin in the gut. If melatonin is produced in the mitochondria, that means it is produced throughout the body and that points to the significance and importance of melatonin in human health. Add in the fact that melatonin can go just about anywhere in the body and the importance of melatonin for health becomes more apparent. The body is loaded with melatonin receptors.

The CVD protective and anticancer effects are just two facets of the multifaceted smart molecule that melatonin is.

This may be the reason why Ayurveda suggests to refrain from eating after sunset.

Maybe intracellular melatonin does not interfere with insulin, but pineal based circulating melatonin does?
