"In conclusion KSM-66, W. somnifera root extract, demonstrated neuroprotective effects showing a capacity to increase the viability of SH-SY5Y cells, a Parkinson's Disease model, to increase glutathione peroxidase and thiol transferase enzyme activities, to modulate the expression of oxidative stress response proteins; peroxiredoxin I, VGF and vimentin, to increase intracellular ATP levels and to modulate redox regulation by decreasing glutathionylated protein levels. These results suggest that KSM-66 is a promising lead drug target for modulating cell damage and cell death due to oxidative stress. Further studies to elucidate the KSM-66 active components and the mechanisms of action are required."

"Results: Research reports based largely on preclinical studies as well as few clinical trials have highlighted the neuroprotective role of Ashwagandha against many neurodegenerative diseases including Alzheimer's, Huntington's and Parkinson's disease. The protective effects of Ashwagandha were accomplished by restoring mitochondrial and endothelial function, mitigation of apoptosis, inflammation and oxidative stress mechanisms.
Conclusion: In this review, we recapitulated neuroprotective properties of Ashwagandha extracts and/or its major constituents and discussed their mechanisms of action and potential therapeutic applications. The pre-clinical as well as clinical studies suggest the use of Withania somnifera (L.) against neurodegenerative disease. However, extensive studies are warranted to validate the use of extract or its single constituents for its clinical use. "

"Abstract: Parkinson’s disease (PD) is characterized by progressive neurodegeneration in the substantia nigra (SN) region resulting in loss of movement coordination. Current therapies only provide symptomatic relief, and there is no agent to halt the progression of PD. Previously, Ubisol-Q10, a water-soluble formulation of coenzyme-Q10, and ethanolic root extract of ashwagandha (ASH) have been shown to inhibit PD pathology in rodent models when used alone. Here, we evaluated the neuroprotective efficacy of oral administration of ASH and Ubisol-Q10 alone and in combination in a paraquat-induced PD rat model. The combined treatment resulted in better-preserved neuron morphology compared to Ubsiol-Q10 or ASH alone. The combination treatment enhanced activation of pro-survival astroglia and inhibited pro-inflammatory microglia. While anti-oxidative effects were seen with both agents, Ubisol-Q10 activated autophagy, whereas ashwagandha showed a better anti-inflammatory response. Thus, the combined treatment caused inhibition of oxidative stress, autophagy activation, inhibition of pro-inflammatory microglia, and activation of pro-survival astroglia. Consequently, paraquat (PQ)-treated rats given the combination treatment in drinking water did not show motor impairment. Based on these interesting observations, the combined treatment containing two well-tolerated natural compounds could be a more effective strategy to halt the progression of PD."
So do you take both Ubsiol-Q10 and ashwagandha? If so - in what form? My HWP does ashwagandha tea - but I have considered the gummies. He also takes ubiquinol. Which covers it right? I don't know how to access which would be better the tea or the gummies.
Apologies if I gave the wrong impression, but I don't take Ubsiol-Q10 or Ashwagandha. I used to take Ashwagandha but I have Hashimoto's and read I should not be taking it. I am still researching to see if it would be safe for me.
I take KSM-66. I find it clears my mental fog better than other Ashwagandha supplements. I take 400 mg in the morning because it tends to make me itch and night-time itching is a big no-no. Well not really an itch - more like the feeling something is crawling on me.
