Hello, my mum has cll and is taking imbruvica.

She suspects she took double dose today (3×140mg) twice. And her dr. Lives in another country, so its not easy to reach him. Anyone has experienced that before and cab advice on what to do? Shall she worry? Shall she not take tomorrows dose?

During the clinical trial process, higher doses than the 420mg amount - including double the standard dose eventually approved, were evaluated,: ncbi.nlm.nih.gov/pmc/articl...

plus there's this current clinical trial that includes double the usual dose:

clinicaltrials.gov/ct2/show...

so I wouldn't be too concerned about the possible double dose your mother may have taken. It's also taken at a higher dose in another blood cancer. The Imbruvica website does urge those that have taken a higher dose to 'call your healthcare provider or go to the nearest hospital emergency room right away.'

imbruvica.com/cll/dosing-an...

but given the above, I wouldn't be concerned.

Given the Imbruvica website doesn't give guidance for your mother's situation, she should call her specialist.

i was on 560mg every day for over a year (now on venetoclax). The half life of imbruvica is supposed to be 4-13 hours

4.9 Overdose

There are limited data on the effects of IMBRUVICA overdose. No maximum tolerated dose was reached in the phase 1 study in which patients received up to 12.5 mg/kg/day (1,400 mg/day). In a separate study, one healthy subject who received a dose of 1,680 mg experienced reversible grade 4 hepatic enzyme increases [aspartate aminotransferase (AST) and alanine aminotransferase (ALT)]. There is no specific antidote for IMBRUVICA. Patients who ingested more than the recommended dose should be closely monitored and given appropriate supportive treatment.

I agree with all the above and think perhaps drinking additional water would be helpful for peace of mind.

I think one extra dose isn’t gone to hurt her.

I would get her one of those pill boxes or someone to come in and help her with her meds by filling the box for her.

Get her a calendar and one of those clocks with the time, day and date.

She may be at the age where it’s best that she gets help with her meds.

I would focus how to not let this happen again.
