Piperlongumine inhibits cell growth and enhances TRAIL-induced apoptosis in prostate cancer cells
“Conclusions: Our findings indicate that piperlongumine can sensitize cancer cells to TRAIL through the upregulation of death receptors and can trigger apoptosis with the downregulation of anti- apoptotic proteins.”
What on earth does that mean? No clue what piperlongumine might be but triggering spoptosis sounds good? I’m guessing most of us here have no clue either. Help?

So what commercial product , made by a reliable manufacturer should be used if one wanted to implement this a additional protocol? Also, what dosage of the product matches the amount used in the studies?

Apparently it is a pepper used extensively in Ayurvedic medicine. Studies of it and its' derivatives have shown pleiotropic activity across various epigenetic pathways inhibiting cancer progression, inducing apoptosis, and selectively killing senescent cells.
TRAIL is "The immune cytokine tumor necrosis factor-related apoptosis-inducing ligand"

Basically it seems to be an immunotherapeutic pathway to kill cancer cells. Many cancer cells either have or can develop resistance to TRAIL and piperlongumine is able to lower or eliminate this resistance.

Have you had any side effects? I know when you take turmeric it’s better to take it with pepper because it helps the body to absorb it better. I believe taking piperlongumine can enhance the absorption of many medications so you have to be careful taking it in supplement form. You can just grind the whole dried peppers and use them as you would black pepper. I don’t know how much of it you would need to see the results they are getting in these studies.
Sounds promising but studies only still being done in vitro for its effects in cancer. So many of these studies don’t pan out. Let’s hope this is one that does. In the meantime just use it as pepper and, who knows?
Hi Faith,Like everything else (the studies or product labels) never or seldom tell you there are many variables affecting the efficacy of a supplement or even drug: weight, age, sex constitution in general. I have not had any problems with piper. I have gotten lazy in my dotage: I used to take a form of Curcumin which is supposed to be more assimilable than the products which use piperine. I did this for many years until we (my wife also takes tumeric) started buying the stuff at Costco in bulk whenever they have a sale. I also used to eat raw Habaneras on a slice of buttered toast (fat soluble) in the day!

A major part of positive preclinical research findings are not replicated when tested clinically and this has apparently not been done with piperlongumine. But its hard not to be impressed by the extraordinary diversity in anti-tumor action of piperlongumine, PL, for a variety of cancers including PCa, preclinically.
Very interestingly, PL is a (possible) senolytic agent, along with the more famous fisetin, dasatinib, quercetin.

From "Senolytic activity of iperlongumine analogues: synthesis and biological evaluation" Xingui Liu et al, 2018:,
"PL appears to be very safe, and its maximum tolerated oral dose in mice is >1 g/kg (lack of acute toxicity after 6 days) while the effective therapeutic doses for cancer can be as low as 2.4 mg/kg by oral dosing. Thus, PL was considered as a novel lead for the development of senolytic agents".
A shot in the dark: Say 12 mg/kilo, five times the lowest dose for efficacy in mice, is in the anti-cancer range for humans, if mice studies and efficacy are transferable, a huge unknown. This gives 0.9 gram, 900 mg per day for a 75 kg person. Packets of PL powder come in 50 and 100 g sizes, meaning they would be used in about two and four months respectively. A normal rate of use? 1- 2 teaspoons per day of black pepper, not PL, is regarded in one study as the upper safe limit meaning 3-6 grams.
The Life Extension product mentioned by Kuanyin has only 10 mg of PL per capsule..
PL may cause GI issues and promote the absorption and availability of some drugs, possibly meaning a dangerously high absorption.
So if we take the traditional approach. Add mega vit c. Curcumin quercitin piper longum reseveratol green tea lycopene bean curd ivermectin Rick Simpson oil and melatonin can we cure ourselves?
